摘要
目的:研究蟾毒灵对人膀胱癌T24细胞增殖和凋亡的影响,初步探讨其在膀胱癌中的抗肿瘤机制。方法:采用MTT法测定蟾毒灵对人膀胱癌T24细胞增殖的影响;流式细胞术(FCM)检测蟾毒灵对癌细胞凋亡和细胞周期的影响;RTPCR法检测癌细胞周期蛋白D1(CyclinD1)基因表达情况。结果:蟾毒灵可抑制人膀胱癌T24细胞的增殖,并呈现出时间和浓度依赖性;蟾毒灵能诱导T24细胞凋亡;20nmol/L浓度的蟾毒灵预处理膀胱癌T24细胞后,与对照组(36.42%)进行比较,可分别在12h(69.76%)和24h(88.14%)显著诱导细胞G0/G1期阻滞(P<0.05),且可降低膀胱癌T24细胞的CyclinD1mRNA表达水平。结论:蟾毒灵能显著抑制人膀胱癌T24细胞增殖,诱导T24细胞G0/G1周期阻滞,下调人膀胱癌T24细胞CyclinD1基因表达。
ABSTRACT AIM: To investigate the effect of bufalin on the proliferation of human bladder cancer T24 cells and explore the underlying anti- cancer mechanisms of bufalin. METHODS: The proliferation-inhibiting effect of bufalin on T24 cells was measured by methyl thiazole tetrazoli- um (MTT)colorimetric assay the cell cycle dis- tribution and apoptosis were detected by flow cytometry ( FCM ) the expression of CyclinD1 was detected by RT-PCR. RESULTS:Bufalin in- hibited the proliferation of T24 cells in a time- and dose-dependent manner bufalin could in- duce apoptosis of T24 cell exposure of T24 cells to 20 nmol/L of bufalin led to significant induc-tion of G0/G1 growth arrest at both 12 h (69.76%) and 24 h (88.14%), compared with negative control cells (36.42%). The mRNA levels of CyclinD1 in T24 cells was markedly de- creased after treatment with bufalin. CONCLU- SION: Bufalin could inhibit the proliferation and induce apoptosis of T24 cells. Bufalin could in- duce T24 G0/G1 growth arrest and decrease the mRNA levels of CyclinD1.
