Effect of bone marrow mesenchymal stem cells on lipopolysacharide-induced secretion of inflammatory cytokines in rat macrophages in vitro

摘要 目的探讨骨髓间充质干细胞对脂多糖(LPS)刺激巨噬细胞分泌炎症因子IL-10、TNF-α的影响。方法体外分离、培养、鉴定骨髓间充质干细胞与巨噬细胞,分PBS、BMSCs、Macrophage、Macrophage+BMSC、BMSCs+LPS、Macrophage+LPS、Macrophage+BMSCs+LPS 7组进行细胞共培养实验。收集不同时间点培养基上清液,ELISA检测其炎症因子IL-10、TNF-α的含量。结果 B(BMSCs)组与E(BMSCs+LPS)组相比,IL-10与TNF-α的含量没有明显变化;F(Macrophage+LPS)组IL-10、TNF-α的含量均高于C组(Macrophage);G(Macrophage+BMSCs+LPS)组与F(Macrophage+LPS)组相比,TNF-α明显降低、IL-10明显升高。结论 BMSCs可以减轻LPS对巨噬细胞分泌炎症因子的刺激作用,减轻炎症反应的程度。 Objective To investigate the effect of bone marrow mesenchymal stem cells (BMSCs) on secretion of inflammatory cytokines in macrophages stimulated by lipopolysacharide (LPS). Methods Rat BMSCs and macrophages were isolated, cultured, and identified. The BMSCs and macrophages, cultured alone or in co-culture, were treated with LPS or PBS or without treatment and tested for interleukin-10 (IL-10) and tumor necrosis factor-a (TNF-a) concentrations in the supernatants at 1, 3, 5, 7, 12, and 24 h after the treatment using enzyme-linked immunosorbent assay. Results Exposure to LPS caused significantly increased IL-10 and TNF-a concentrations in the supernatant of cultured macrophages but not in BMSC culture. Macrophages co-cultured with BMSCs showed significantly lowered IL-10 and TNF-a secretions in response to LPS exposure as compared with the macrophages cultured alone. Conclusion BMSCs can reduce LPS-induced secretion of inflammatory cytokines by the macrophages to ameliorate inflammatory reactions.
