摘要
目的通过建立类风湿性关节炎CIA模型,探讨食物抗原与类风湿性关节炎的相关性,为临床预防和治疗类风湿性关节炎提供理论依据。方法对Wistar大鼠尾根、足部和脊柱两侧注射牛Ⅱ型胶原免疫诱导建立CIA模型,通过组织病理学和免疫学对模型进行评价,检测TNF-α、IL-17和IL-1细胞因子的表达水平;通过酶联免疫吸附法(ELISA)和免疫印迹法(Western blot)分析鸡蛋和牛奶粗提抗原与CIA模型的相关性。结果 45 d后成功建立CIA模型,ELISA检测发现鸡蛋和牛奶粗提抗原与CIA模型组免疫反应较高,Western blot检测到鸡蛋和牛奶与CIA模型组免疫反应存在特异性蛋白,蛋白相对分子质量分别在45 000-60 000和80 000-90 000之间。结论食物抗原与类风湿性关节炎存在一定的相关性,鸡蛋和牛奶中的特异蛋白的种类及作用需进一步研究。
This study designed to explore the relevance between food antigens and RA by establishing rat model of collagen-induced arthritis(CIA), with the aim of providing theoretical basis for clinic prevention and treatment of RA. Firstly, CIA model was established by injecting Bovine Collagen type II at four points of the wistar rat, namely the tail, foot and two sides of instep, and then confirmed by means of histopathology and immunology.Furthermore, we evaluate the rheumatoid inflammation in CIA model by detecting the expression of TNF-α, IL-17 and IL-1. Then the correlation between RA and the crude extracts of egg and milk was evaluated using means of ELISA and Western blot. Data showed the antigens in egg and milk extracts possessed higher immunogenicity, and had intersect immune reaction with CIA. Western blot indicated that there were special proteins in egg and milk,with molecular mass of 45-60 kD and 80-90 kD, respectively. All result showed that food allergens is related with RA. And we initially believe there are two kinds of proteins in egg and milk correlated with CIA, but the concrete detail needs further study.
