三七总皂甙对肝癌细胞株、甲胎蛋白异质体及高尔基体糖蛋白-73表达的影响 被引量:1

摘要 目的:研究三七总皂甙(panax notoginsenosides,P N S)对不同肝癌细胞株、甲胎蛋白异质体(alpha-fetoprotein-L3,AFP-L3)分泌及高尔基体糖蛋白-73(Golgi glucoprotein 73,GP-73)的影响.方法:以肝癌细胞HepG2两种细胞株SMMC-7721和Bel-7402作为研究对象,分为空白对照组、5-氟尿嘧啶(5-fluorouracil,5-Fu)组和不同浓度梯度(10-400μg/mL)的PNS组,共培养120 h,利用MTT比色测定法检测细胞活性,ELISA及细胞分化法检测AFP-L3及GP-73活性.结果:PNS在>200μg/mL时可表现出对两种肝癌细胞株增殖的抑制,高浓度PNS(400μg/mL)对细胞活性影响较大,抑制率与5-Fu(40μg/mL)的作用相似.PNS组200、400μg/mL剂量均明显抑制肝癌两种细胞株GP-73活性,且剂量400μg/mL的抑制作用与5-Fu组比较有显著差异(P<0.05).PNS组剂量达400μg/mL时才能有效抑制肝癌两种细胞株AFP-L3活性,与对照组比较差异显著(P<0.01);且400 ng/mL剂量的抑制作用与5-Fu组没有显著差异.结论:PNS具有抑制肝癌细胞株的生长,降低AFP-L3与GP-73活性,肝癌细胞基因分化,诱导其在体外向正常肝细胞分化,且效果与剂量-时间呈正相关. AIM: To study the effect of panax notoginsen-osides(PNS) on the expression of alpha-feto-protein-L3(AFP-L3) and Golgi glucoprotein 73(GP-73) in hepatoma cell lines. METHODS: Hepatoma cell lines SMMC-7721 and Bel-7402 were used. The cells were dividedinto different groups and treated with 5-fluoro-uracil(5-Fu) or different concentrations of PNS(10-400 μg/mL) for 120 h. Untreated cells wereused as controls. MTT assay was used to detectcell activity, and enzyme linked immunosorbentassay(ELISA) was used to detect AFP-L3 andGP-73 levels. RESULTS: PNS at concentrations 200 μg/mLshowed a significant inhibitory effect on hepa-toma cell growth. PNS at a concentration of400 μg/mL had comparable inhibitory effect to 5-Fu at 40 μg/mL. PNS at concentrations of both 200 and 400 μg/mL significantly inhibited GP-73 activity in hepatoma cells, and the in-hibitory effect of PNS at 400 μg/mL was signifi-cantly higher than that of 5-Fu at 40 μg/mL(P〈0.05). PNS at 400 μg/mL could significantly suppress AFP-L3 in hepatoma cells compared with controls(P〈0.01), and the effect was sim-ilar to that of 5-Fu. CONCLUSION: PNS can inhibit hepatoma cell proliferation and suppress AFP-L3 and GP-73 activity.
