摘要
[目的]研究离子液体1-乙基-3-甲基咪唑磷酸二乙酯磷酸盐(〔Emim〕DEP)对酿酒酵母SY101生长的影响。[方法]用显微镜观察了不同〔Emim〕DEP浓度下液体培养基中对数生长期酵母细胞的形态及出芽情况,用分光光度法测得的数值绘制出酵母菌24 h生长曲线。[结果]试验发现,〔Emim〕DEP对酿酒酵母SY101生长有抑制性,随着〔Emim〕DEP浓度的增加,抑制作用越明显。当〔Emim〕DEP浓度高于3 g/L时,酵母菌的形态发生明显改变;浓度高于10 g/L时,酵母菌的出芽率明显下降。通过平板培养测得〔Emim〕DEP对酵母菌的半有效浓度EC50值为6.67 g/L。[结论]研究可为由离子液体从农林废弃物中制备纳米纤维素后的残渣发酵燃料酒精的研究提供基础数据。
The effects of ionic liquid 1-ethyl-3-methylimidazolium diethylphosphate(〔Emim〕DEP) on growth activity of Saccharomyces cerevisiae SY101 were studied by signal factor tests. [Method]Observed the morphological structure and budding of Saccharomyces cerevisiae SY101 at Log growth phase in the presence of different concentration of〔Emim〕DEP. Drawn growth curve according to numerical value by spectrophotography in liquid suspension culture. [Result]The results indicated that〔Emim〕DEP could inhibit the growth of yeast. The higher the〔Emim〕DEP concentration,the more obvious the inhibition effect was. The yeast's single cell morphology changed when〔Emim〕DEP concentration was greater than 3 g /L,and its reproduction rate by budding decreased when〔Emim〕DEP concentration was greater than 10 g /L. The half effective concentration(EC50) of〔Emim〕DEP to Saccharomyces cerevisiae SY101 were measured by using solid and value were 6.67 g/L.[Conclusion]The study can provide basic data for fuel alcohol residue fermentation.
