摘要
目的观察松郁安神方对睡眠剥夺大鼠血清细胞因子的影响,进一步探讨松郁安神方调节睡眠的机制。方法采用"改良多平台睡眠剥夺法"构建睡眠剥夺大鼠模型,以正常笼养及大平台作为对照组,采用酶联免疫吸附法测定睡眠剥夺大鼠经松郁安神方治疗后,血清细胞因子IL-1β、TNF-α水平的变化。结果松郁安神方组血清TNF-α高于REM睡眠剥夺组(P<0.05);松郁安神方组血清IL-1β较正常对照组、大平台环境对照组及REM睡眠剥夺组无显著性差异(P>0.05)。结论松郁安神方可能通过调节血清细胞因子TNF-α的含量从而调节睡眠,且通过对睡眠剥夺大鼠睡眠的调节继而改善睡眠剥夺所致机体的过度能量代谢。
OBJECTIVE To observe the effects of SongYuAnShenFang on serum cytokines levels of sleep depri- vation rats. METHODS The sleep deprivation in rats model was constructed with the method called "modified mul- tiple platform method". Comparions were housed either in normal cages or in tanks with large platforms. After the treatment of SongYuAnShenFang in Sleep Deprivation Rats, serum levels of IL-1β ,TNF-α were measured by enzyme linked immunosorbent assay. RESULTS Significant differences in serum level of TNF- αwere observed between SongYuAnShenFang group and REM sleep deprivation group( P 〈0.05 ). There was no significant differences of serum level of IL-1β in SongYuAnShenFang group compared with the other three groups ( P 〉 0. 05 ). CONCLUSION SongYuAnShenFang in improving sleep quality may be based on its regulation of serum level of TNF-ct, then adjust the excessive body metabolism caused by sleep depriveation.
