摘要
目的建立走马胎中多糖含量测定的方法,并测定走马胎中多糖的含量,通过单因素考察对其提取工艺进行初步研究。方法采用苯酚-硫酸法于488 nm波长处测定多糖的含量。考察因素有提取时间、提取次数、料液比。结果在拟订的试验条件下,走马胎的多糖含量为1.15%,平均回收率为101.22%,RSD=2.32%(n=6)。所得最佳单因素分别为水提时的提取时间2.5 h,提取次数1次,料液比1:30。结论苯酚-硫酸法测定走马胎中多糖,方法简便、准确、稳定性好。
Objective To establish the determination method of polysaccharide content of Ardisia Gigantifolia Stapf and to perform the preliminary study on its extraction process. Methods The phenol - sulfuric acid method was adopted to detect the polysaccharide content at the wavelength of 488 nm. The investigation factors included the extraction time, extraction times and the solid- liquid ratio. Results In the proposed experimental conditions, the polysaccharide content of Ardisia Gigantifolia Stapf was 1.15%, the average recov- ery rate was 101.22%, RSD=2.32%. The obtained best single-factor was that in the water extraction, the extraction time was 2.5 h, extracting once, the solid- liquid ratio was 1:30. Conclusion The phenol- sulfuric acid method for determining the polysaccharide content of Ardisia Gigantifolia Stapf is simple and accurate with good stability.
