摘要
目的:建立大鼠血浆中木蝴蝶提取物槲皮素含量的分析方法及药代动力学参数。方法:大鼠灌胃给药10 mg/kg后,于不同时间点眼眶取血;采用HPLC法XDB-C18色谱柱,流动相:乙腈-0.1%冰醋酸水溶液进行梯度冼脱,流速:1.00 mL/min,柱温:30℃,检测波长为:278 nm。分析大鼠血浆样品中槲皮素的含量并计算其药动学参数。结果:槲皮素在0.5~22.5μg/mL范围线性关系良好,r=0.9969,提取回收率83.9%~88.5%。日内日间精密度RSD〈10.0%。10 mg/kg木蝴蝶提取物槲皮素的tmax为(15.60±4.65)min,Cmax为(0.75±0.12)μg/mL,t1/2为(215.5±102.3)min。结论:该法灵敏度高,操作简便、准确,可用于木蝴蝶提取物大鼠灌胃给药后黄酮类成分的测定及药代动力学研究。
Objective: To establish a method of analyzing the content and pharmacokinetic parameters of quereetin of orxylum indicum extracts in rat plasma. Method : After being delivered with diverse doses of 10,20 and 30 mg/kg by intragastric administration. The samples of blood were obtained from rat orbits at different time, respectively. HPLC was carried out to analyze the content of quercetin in rat plasma, which was performed on Agilent Eclipse XDB-C18 column by using a mobile phase of acetonitrile and water-0.1% glacial acetic acid for gradient eluted at a column temperature of 30 ℃ and the detection wavelength was set at 278 nm. And then the pharmacokinetic parameters of quercetin were obtained. Results : Within the range of 0.5-22.5 μ g/mL for quercetin, a good linear relationship was presented, r=0.9969, and the recovery located between 83.9% and 88.5%. The precision RSDs of intraday and interday were less than 10.0%. The tmax of quercetin of extracts with the doses of 10,20, and 30 mg/kg were expressed at ( 15.60 ±4.65 ) min, ( 16.45 ± 5.84 )min and ( 17.36 ± 6.12 )rain, respectively. C showed with the concentrations of ( 0.75 ± 0.12 ) mg/L, ( 1.25 ±0.32 ) mg/L and ( 1.67 ± 1.25 ) mg/L, and the T1/2 obtained at ( 215.5 ± 102.3 )min, ( 220.5 ±110.2 )min and ( 218.5 ± 106.5 )rain. Conclusion: The method has the advantages of high sensitiveness, simple operation and accuracy, which can be applied for the determination of the content and the investigation of pharmacokinetics of flavonoids of orxylum indicum extracts after intragastric administration for rat.
