摘要
目的分析SLE患者外周血单个核细胞(PBMC)中微小RNA(miR)-132和miR-212的表达水平及临床意义。方法提取PBMC中的总RNA,运用茎环逆转录实时荧光定量PCR(qRT-PCR)技术检测24例健康人及48例SLE患者PBMC中miR-132和miR-212的相对表达水平,分析其与临床检验指标的相关性。结果 SLE患者中miR-132相对表达量为1.72±0.071,与健康人对照组0.99±0.037比较,差异有统计学意义(t=-6.881,P=0.000)。健康人和SLE患者miR-212相对表达水平分别是0.99±0.020和1.03(0.982,1.048),差异未见统计学意义(Z=1.302,P=0.197)。相关性分析结果显示,SLE患者中miR-132的表达量与抗双链DNA(dsDNA)抗体以及SLE患者疾病活动指数(SLEDAI)呈正相关(r分别为0.740和0.526,P均<0.05);而miR-212的表达量与抗dsDNA抗体(r=0.106,P=0.474)和SLEDAI(r=0.005,P=0.973)等临床指标无相关性。结论 miR-132在SLE患者PBMC中的表达水平升高,可能参与SLE的致病过程。
Objective To investigate the expression levels of miR-132 and miR-212 in peripheral blood mononuclear cells( PBMCs) of patients with systemic lupus erythematosus( SLE) and their clinical significance. Methods Total RNAs were extracted from PBMCs of48 SLE patients and 24 healthy controls( HC). Then,the relative expression levels of miR-132 and miR-212 were detected by stem-loop reverse transcription quantitative real time polymerase chain reaction( qRT-PCR),and their correlations with clinical parameters were analyzed. Results The relative expression level of miR-132 in SLE patients( 1. 72 ± 0. 071) was significantly higher than that in HC( 0.99 ±0.037,t =-6.881,P〈0.001). However,there was no any significant difference in miR-212 between SLE patients and HC( 1.01 ±0.068 vs 0.99 ±0.020,Z =1.302,P =0.197). The expression level of miR-132 in SLE patients was positively related to antidsDNA concentration and systemic lupus erythematosus disease activity index( SLEDAI)( r = 0. 740 and 0. 526,P〈0. 05),but the miR-212 level unrelated to them( r = 0. 106,P = 0. 474 and r = 0. 005,P = 0. 973). Conclusion The expression level of miR-132 in PBMCs of SLE patients increases,indicating that miR-132 may play an important role in the development and progression of SLE.
