摘要
目的:建立同时测定旱莲草提取物中木犀草素和芹菜素含量的方法。方法:采用高效液相色谱法。色谱柱为Nucleodur100-5 C18(250 mm×4.6 mm,5μm),流动相为甲醇-0.2%磷酸水溶液(50∶50,V/V),流速为1 ml/min,检测波长为350 nm,柱温为40℃结果:木犀草素和芹菜素的质量浓度均在0.02-50.00μg/ml范围内与各自峰面积积分值呈良好的线性关系(r分别为0.9995和0.9993);精密度、稳定性、重复性试验的RSD〈2%;平均加样回收率分别为97.2%和98.9%,RSD分别为2.41%和2.91%(n均为6)。结论:该方法快速、简便,结果准确,可用于不同产地旱莲草药材的质量评价。
OBJECTIVE:To establish a method for simultaneous determination of luteolin and apigenin in Eclipta prostrata extract. METHODS:HPLC method was adopted. The separation was carried out on Neocleodur 100-5 C18(250 mm×4.6 mm,5 μm)column with mobile phase consisted of methanol-0.2% phosphoric acid(50∶50,V/V)at the flow rate of 1 ml/min. The detection wavelength was set at 350 nm and the column temperature was maintained at 40 ℃ RESULTS:The linear range were 0.25-50.00μg/ml for luteolin(r=0.999 5)and 0.25-50.00 μg/ml for apigenin(r=0.999 3);RSDs of precision,stability and reproducibility tests were lower than 2%. Average recovery rates were 97.2%(RSD=2.41%,n=6)and 98.9%(RSD=2.91%,n=6),respectively. CONCLUSIONS:The method is rapid,simple and accurate,and can be used for the quality evaluation of E. prostrata.
