新疆药桑叶的HPLC指纹图谱研究

Study on HPLC Fingerprint of Morus alba Leaves from Xinjiang

目的:建立新疆药桑叶的高效液相色谱(HPLC)指纹图谱。方法:采用HPLC法。色谱柱为YMC-C18(250 mm×4.6 mm,5μm),流动相为0.1%磷酸水溶液-0.2%磷酸乙腈溶液(梯度洗脱),检测波长为360 nm,流速为1.0 ml/min,柱温为30△,进样量为10μl。以异槲皮苷为参照物,分析10批次不同产地的药桑叶样品,采用计算机辅助相似度评价系统进行评价。结果:药桑叶指纹图谱由11个特征峰构成,10批次样品指纹图谱的相似度均>0.90,不同产地药桑叶的主成分组成基本相同。结论:所建立的指纹图谱稳定性、重复性好,可用于新疆药桑叶的鉴别和质量控制。

OBJECTIVE：To establish HPLC fingerprint of Morus alba leaves from Xinjiang. METHODS：HPLC method was adopted. The determination was performed on YMC-C18（250 mm×4.6 mm,5 μm）column with mobile phase consisted of 0.1%phosphoric acid solution-0.2% phosphoric acid acetonitrilem solution（gradient elution）at the flow rate of 1.0 ml/min. The detection wavelength was set at 360 nm,and column temperature was 30 ℃. The sample size was 10 μl.10 batches of sample from different origin were analyzed with isoquercitrin as the reference. Computer-aided evaluation system of similarity was used to evaluate the data. RESULTS：The fingerprints constituted by 11 characteristic peaks,and the similarity of HPLC fingerprints of 10 batches of sample were greater than 0.90. The main ingredients of M. alba leaves from different origin were the same basically. CONCLUSIONS：The established fingerprint is stable and reproducible,and can be used for the identification and the quality control of M.alba leaves.
