摘要
采用高相液相色谱-质谱联用技术和偏最小二乘分析(PLS-DA)方法,观察给予半夏厚朴汤水煎剂后大鼠尿样代谢物组的变化;并对抗抑郁机制进行初步解释。实验研究过程中,给药组灌胃给予半夏厚朴汤水提物,借助高效液相-质谱联用仪(LC-MS)检测尿样,收集数据后,采用PLS-DA方法对大鼠尿样内源性小分子代谢产物进行检测和分类处理。结果:三组大鼠代谢特征有显著差异。根据PLS-DA分析的载荷图和VIP值,进一步筛选对数据分类存在较大影响的离子,鉴别得到12个代谢标记物。结果是相对于模型组的参与能量代谢的柠檬酸,乳酸,2-酮戊二酸含量减少以及马尿酸,肌酐酸水平升高,给药组在一定程度上可以逆转这种异常改变。结合大鼠体重和行为学结果,表明半夏厚朴汤具有抗抑郁样作用。
Partial Least Squares analysis methods were used to characterize rat urine obtained after chromic exposure Banxia-Houpo decoction. In our study, the treatment group treated with Banxia-Houpo decoction. The principal component analysis method was used to predict and classify treatment of these three groups. When the data were collected, liquid chromatography-mass spectral regions, and related metabolites were identified and ranked by their weight aline with the loading plot. It was not difficult to find that there were obvious differences among the treatment group, the model group and the control group. Loading plot and variable importance in the projection (VIP) of PLS-DA were used to search for the ions which had a significant contribution to classification, and the 12 of these ions were identified. The results showed that decreased levels of lactate, 2-oxoglutaric acid, and elevated levels of creatinine and hippurate in the model group urine, abnormal reversed by the treatment group to some extent , combined with the weight and behavioral science results in rats, suggesting Banxia-houpu decoction has an antidepressant-like effect.
