HPLC-ELSD法测定加味归芪片中黄芪甲苷的含量

Determination of Astragaloside in Modified Guiqi Tablets by HPLC-ELSD

目的应用HPLC-ELSD法测定加味归芪片中黄芪甲苷的含量。方法 HPLC-ELSD法。色谱柱为Waters Fire C18柱（4.6mm×250mm,5μm）;流动相：乙腈-水（32∶68）;流速1.0mL·min-1;柱温：35℃;ELSD参数：雾化器温度40℃,漂移管温度为90℃,气体压力为35psi。结果黄芪甲苷进样量在1.536-61.44μg范围内线性关系良好（r=0.9998,n=8）,平均加样回收率为99.13%,RSD为2.07%,（n=9）。结论该法简便快速,重复性好,灵敏度高,结果准确,可用于加味归芪片中黄芪甲苷含量的测定。

OBJECTIVE To estiblish a HPLC-ELSD method to determin the Astragaloside in medified Guiqi Tablets. METHODS HPLC-ELSD method was used. The column was Waters Fire C18column（ 4. 6mm × 250 mm,5μm）; mobile phase： acetonitrile water was 32∶68; the flow rate was 1. 0mL·min- 1; column temperature was 35℃;ELSD parameter was atomizer temperature of 40℃,and the drift tube temperature of 90℃,gas pressure was35 psi. RESULTS Astragaloside IV has a good linear relationship in 1. 536 ~ 61. 44μg（ r = 0. 9998,n = 8）,the average recovery was 99. 13%,RSD was 2. 07%（ n = 9）. CONCLUSION This method is simple,fast,good reproducibility,high sensitivity,accurate,and can be used for measuring the Guiqi film content of Astragaloside.
