羟基红花黄色素A对急性脊髓损伤模型大鼠神经细胞凋亡的影响 被引量:5

Effects of Hydroxysafflor Yellow A on Neural Cell Apoptosis After Acute Spinal Cord Injury in Rats

摘要 目的观察羟基红花黄色素A(HSYA)对急性脊髓损伤(ASCI)模型大鼠神经细胞凋亡的影响。方法 30只SD大鼠随机分为空白对照组、损伤对照组和HSYA治疗组,每组10只。采用Allens’s打击法建立急性脊髓损伤大鼠模型,分别于术后1、7、14天观察各组大鼠行为学评分,术后14天通过免疫组化方法观察脊髓损伤部位神经细胞caspase-3 p20、Bcl-2和Bax表达。结果HSYA治疗组与损伤对照组大鼠下肢功能均有恢复(P约0.05),且HSYA治疗组恢复更明显(P约0.05);与损伤对照组比较,HSYA治疗组caspsase-3p20阳性细胞数表达明显减少(P约0.05),Bcl-2阳性细胞显著增加(P约0.05),Bax阳性细胞明显减少(P约0.05)。结论 HSYA对急性脊髓损伤模型大鼠神经细胞凋亡有明显抑制作用,从而减少脊髓损伤部位继发性损伤,促进功能恢复。 Objective To determine the therapeutic efficacy of hydroxysafflor yellow A (HSYA) treatment on neural cell apoptosis after acute spinal cord injury(ASCI) in rats. Methods ASCI was induced by using the mod-ified weight-drop method in Sprague-Dawley rats. The ASCI rats were randomly divided into three groups:sham group(laminectomy only,n=10),vehicle-treated group(n=10),and HSYA-treated group(n=10). Locomotor functional recovery was assessed by performing open-field locomotors tests on postoperative day 1,7,and 14. On postoperative day 14,segments of spinal cord encompassing the injury site were removed and collected for histopathological anal-ysis. Immunohistochemistry was performed to observe the expression of B-cell CLL/lymphoma-2(Bcl-2),BCL-2-as-sociated X protein (Bax) and caspase-3 p20. Results Rats in HSYA-treated group and vehicle-treated group had significantly better locomotor function recovery than those in sham group (P〈0.05),and the difference between HSYA-treated group and vehicle-treated group was also significant (P〈0.05). Compared with vehicle-treated group, spinal tissues from HSYA-treated rats had increased expression of Bcl-2 (P〈0.05) and reduced expression of cas-pase-3(P〈0.05) and Bax(P〈0.05) after spinal cord injury. Conclusion HSYA treatment after SCI can significantly improve locomotor recovery, and this neuroprotective effect may be related to inhibition of neural apoptosis.
