分散液液微萃取/荧光分光光度法快速测定乳制品中的黄曲霉毒素M_1 被引量:11

Determination of Aflatoxin in Dairy Products by Dispersive Liquid-Liquid Microextraction and Fluorescence Spectrophotometry

摘要 基于分散液液微萃取技术和荧光增强技术,建立了简便、快速、经济的分散液液微萃取/荧光光度法用于测定乳制品中的黄曲霉毒素M1(AflatoxinM1,AFM1)。以二氯甲烷作为萃取剂,乙腈作为分散剂,用荧光分光光度计测定萃取液。实验结果表明,AFM1在0.05~2.00μg/L范围内线性关系良好,对液态奶和固态奶粉两类乳制品的相关系数(r)分别为0.9956和0.9989,检出限(LOD)分别为0.023μg/L和0.039μg/L,样品加标回收率为76.7%~95.7%,相对标准偏差(RSD,n=6)小于5%。经统计分析,该法与国标标准方法的准确度无显著差异,可应用于奶制品中AFM1残留量的快速检测。 A simple, rapid, and low-cost method based on dispersive liquid - liquid microextraction (DLLME) and fluorescence enhancement spectrophotometry was developed to detect aflatoxin M1 (AFM1 ) in dairy products. Methylene chloride( CH2C12 ) and acetonitrile were used as extracting agent and dispersant, respectively. The extract was detected by fluorescence spectrophotometry. Under the optimum conditions, good linear relationships were obtained for AFMl in the concentration range of 0. 05 -2.00 μg/L with correlation coefficients(r) of 0.995 6(milk) and 0.998 9( milk powder). The limits of detection(LOD) were 0.023 μg/L for milk and 0.039 μg/L for milk powder. The spiked recoveries were in the range of 76.7% - 95.7% with relative standard deviations ( n = 6) lower than 5% . The method could be applied in fast detection of aflatoxin M1 in dairy products, and there is no significant difference between this method and the national standard method.
