芪附汤对心肌缺血再灌注损伤大鼠心电图和血流动力学变化的影响 被引量:4

摘要 目的:观察芪附汤对大鼠心肌缺血再灌注损伤后不同时间点的心电图、血流动力学等相关指标的影响,探讨芪附汤对大鼠心肌缺血再灌注损伤的药效作用.方法:SD大鼠30只,随机分为5组:空白组、模型组、阳性组(盐酸维拉帕米片)4mg/kg、芪附汤高剂量组1.2g(生药)/kg、芪附汤低剂量组0.6g(生药)/kg,各组灌胃给药,连续5天,末次给药30min后,结扎大鼠心脏冠状动脉左前降支复制心肌缺血再灌注模型,Power-lab生理记录仪描记心电图、血流动力学曲线,观察造模后各个时间点心电图ST值和血流动力学相关指标的变化.结果:空白组与模型组比较,大鼠心肌缺血再灌注后10~30min,心电图ST段抬高值下降大于50%,表明造模成功.芪附汤高、低剂量给药组大鼠与模型组大鼠比较,造模后1h能显著性降低ST抬高值(P<0.01),造模后2h能显著升高大鼠左心室收缩压(LVSP)的降低值(P<0.05),造模后1h均能显著升高左室内压最大上升速率(+ dp/dt max)的降低值(P<0.01),造模后2h均能显著降低左室内压最大下降速率(-dp/dt max)的抬高值(P<0.05),造模后2h和3h均能显著升高大鼠舒张压(DAP)的降低值(P<0.05);造模后2h,芪附汤高给药组大鼠能显著升高大鼠收缩压(SAP)、平均动脉压(MAP)的降低值(P<0.05).结论:芪附汤对大鼠心肌缺血再灌注损伤有显著的保护作用,并呈现一定的药效作用规律,为其临床治疗冠心病、心绞痛等提供实验依据. Objective : To observe the effect of Qifu Decoction on myocardial ischemia reperfusion injury in rats after different time points of electrocardiogram ( ECG), blood flow dynamics and other related indicators, To study the effect of Qifu Decoction on myocardial is- chemia- reperfusion injury in rats. Methods: 30 SD rats were randomly divided into 5 groups:sham -operation group, model group, Verapamil Hydrochloride Tablets (4mg/kg), the large dose groups of Qifu Decoction 1.2g( crude drug)/kg, the low dose groups of Qifu Decoction 0.6g ( crude drug)/kg, Each group was ig administrted for 5 consecutive days. The model of myocardial ischemia reper- fusion injury in rats was induced by ligation of the left anterior descending of the coronary artery (LAD) after 30 minutes of end admin- istration. Electrocardiogram (ECG), Hemodynamic curve was recorded by The Power- lab physiological grapher, Each time point ST value changes of ECG were observed and hemodynamic indexes. Result: Compared the blank group and the model group, After myo- cardial ischemia - reperfusion 10 to 30 min in rats,The increase values of Ecg ST segment drop more than 50% ,That building success. Qifu Decoction of high and low doses in rats compared with model group rats, building 1 h after ST elevation values have dropped sig- nificantly ( P 〈 O. 01 ), 2 h after building can marked increase in rats with left ventricular systolic pressure (LVSP) ( P 〈 0.05 ), lower value of building 1 h after all can the marked increase in left ventricular isovolumetric systolic interior maximum pressure rise rate ( + dp/dt Max) of lower value (P 〈 0. O1 ), 2 h after building can significantly reduce the left ventricular isovolumetric diastolic pressure reduced rate of the largest indoor ( + dp/dt Max) of higher value (P 〈 0.05), 2 h and 3 h after building are to a significant rise in di- astolic blood pressure of rats (DAP) of lower value (P 〈 0.05 ) ; Building after 2 h, stilbene decoction high dose group of rats signifi- cantly elevated systolic blood pressure (SAP) in rats, reduce the value of the mean arterial pressure (MAP) ( P 〈 0.05). Conclusion : Qifu decoction has a obviousprotective effect on myocardial ischemia -reperfusion injury. And Render certain therapeutic effects rule, so provide the experimental evidence its clinical treatment of coronary disease, angina pectoris etc.
