摘要
目的:研究人参二苓汤对H22荷瘤小鼠恶性腹水的抑制作用及相关机制。方法:建立H22荷瘤小鼠恶性腹水模型,随机分为模型对照组,阳性对照组,人参二苓汤有效部位低、中、高剂量组,观察各组小鼠毛色、食欲、活动状况和精神状态等一般的生活状况;测定小鼠体重和腹围的变化;观察腹腔内情况,计数腹膜瘤结节数量,称重计算抑瘤率;利用Elisa法检测腹水中VEGF、MMP-2、MMP-9的表达量。结果:人参二苓汤有效部位能够延缓H22荷瘤小鼠体重和腹围的增长,对H22荷瘤小鼠腹水的生长有一定的抑制作用,且存在剂量依耐性。与模型组比较,人参二苓汤有效部位高剂量组小鼠腹水中VEGF的含量显著降低(P<0.01),MMP-2和MMP-9的含量有差异(P<0.05);中剂量组小鼠腹水中VEGF的含量有差异(P<0.05),MMP-2和MMP-9的含量无差异(P>0.05);低剂量组小鼠腹水中VEGF、MMP-2和MMP-9的含量均无差异(P>0.05)。人参二苓汤有效部位各剂量组与模型对照组比较均无差异。结论:人参二苓汤能够提高H22腹水型小鼠的生活质量,抑制恶性腹水的生长,抑制腹水中VEGF、MMP-2和MMP-9的表达,而对H22荷瘤小鼠恶性腹水的发展起抑制作用。
Objective : To investigate the inhibitory effect of Renshen Erling Decoction on H22 malignant ascites in mice and the correla- ted mechanisms. Methods: Firstly, build up the model of H22 malignant ascites in mice. Then the mice were randomly divided into the model group, the positive group and the effective part of Renshen Erling Decoction low, medium and high dose group. Finally observe the coat color, appetite, activity, mental state and other general living conditions of each group, measure changes in weight and ab- dorminal perimeter, observe abdorminal conditions including counting the number of peritoneal tumor modules and detect the contents of VEGF,MMP- 2 and MMP- 9 in ascites with Elisa assay. Results: The effective part of Renshen Erling Decoction can delay the growth of weight and abdominal perimeter of H22 tumor - bearing mice and has a certain inhibitory effect with dose - dependent for the growth of ascites. Compared with the model group, the content of VEGF in ascites of the effective part of Renshen Erling Decoction high dose group was significantly decreased (P 〈 O. 01 ), and both of the contents of MMP -2 and MMP -9 were different( P 〈 O. 05 ) ; in the medium group, the content of VEGF was different(P 〈 0.05 ), but both of the content of MMP - 2 and MMP - 9 had no differ- ence ( P 〉 0.05 ) ; however, in the low group, the content of VEGF, MMP - 2 and MMP - 9 showed no difference ( P 〉 0.05 ), and each group of Renshenerling Decoction and the model group had no difference, too. Conclusion: Renshenerling Decoction could improve the living quanlity of H22 tumor - bearing mice, inhibit the growth of malignant ascites, reduce the expression of VEGF, MMP - 2 and MMP -9 in ascites, and suppress the development of H22 with ascites type in mice .
