重金属镉胁迫对日本蟳抗氧化酶活力、丙二醛含量及组织蓄积的影响 被引量:8

Effects of Cadmium Stress on Activities of Antioxidant Enzymes,Malonaldehyde Content and Cadmium Accumulation in Asian Swimming Crab Charybdis japonica

摘要 在15℃下,将体质量(101.53±16.48)g的日本蟳暴露于对照及低质量浓度组(0.0025、0.005mg/L)、中质量浓度组(0.05、0.1mg/L)和高质量浓度组(0.25、0.5mg/L)的氯化镉中,于处理0、1、3、5、7、10、15d和30d时取血,测定其血清超氧化物歧化酶和过氧化氢酶活力、丙二醛含量;并于胁迫第30d,测定肝胰腺、肌肉、卵巢、鳃中的镉含量,以研究镉胁迫对日本蟳抗氧化能力及组织蓄积的影响。结果表明:在镉胁迫下,日本蟳超氧化物歧化酶和过氧化氢酶活力均呈先升后降趋势。中质量浓度组及高质量浓度组的镉胁迫对抗氧化酶活力诱导较快,而低质量浓度组诱导慢。胁迫第15d和30d,除0.0025mg/L组外,试验组两种酶活力均显著低于对照组,与镉胁迫质量浓度呈显著负相关(P<0.05)。0.0025mg/L组丙二醛含量无明显变化,其他各组呈两种变化趋势:0.005、0.05mg/L和0.1mg/L组丙二醛含量先升后降再升,而高质量浓度(0.25、0.5mg/L)组丙二醛含量持续上升。至胁迫第10d,除0.0025mg/L组外,试验组丙二醛含量均显著高于对照组,与镉胁迫质量浓度呈显著正相关。镉在日本蟳肝胰腺和鳃内的积累较多,而卵和肌肉仅在高质量浓度镉胁迫下才有微量的蓄积。各组肝胰腺中镉含量与质量浓度呈明显的质量浓度—效应关系。中质量浓度组和高质量浓度组鳃中镉含量显著增加,超过同一质量浓度组肝胰腺的镉蓄积量。 The activities of superoxide dismutase(SOD)and catalase(CAT),and content of malonaldehyde(MDA)were determined in Asian swimming crab Charybdis japonica exposed to different cadmium(Cd)levels including 0mg/L in control group,0.0025mg/L and 0.005mg/L in LC groups,0.05mg/L and 0.1mg/L in MC groups,and 0.25mg/L and 0.5mg/L in HC groups 0,1,3,5,7,10,15 and 30days after the exposure.The Cd accumulation was detected in the hepatopancreas,muscle,ovary,and gill at the30 th day.The results showed that the SOD and CAT activities were found to be elevated first and then to be declined after Cd exposure,with more rapid induction in MC groups,and HC groups than in LC groups.On days 15 and 30,the activities of SOD and CAT were significantly lower in the treatment groups than those in the control group except in 0.0025mg/L group,with significant negative correlation with Cd concentration(P〈0.05).The MDA contents were first increased,then declined and elevated in the groups with Cd concentrations of 0.005,0.05 and 0.1mg/L.However,the constant increase in MDA contents were observed in HC groups,even though there was no significant change in MDA contents in the 0.0025mg/L group.Until the 10 th day after exposure,the MDA contents were significantly higher in the treatment groups than that in the control,showing apositive correlation with Cd concentration,except in0.0025mg/L group.There was more Cd accumulation in the hepatopancreas and gill,while minute amounts of Cd was detected in the ovary and muscle in the HC groups.Significant effect-dosage relationships were observed between the accumulation content and Cd concentration in the hepatopancreas in the treatment groups.There were more Cd accumulation in the gills than that in hepatopancreas in MC and HC groups.
