摘要 目的:一次性大负荷运动对淋巴细胞线粒体自噬的影响及其生物学效应。方法24名武警战士进行一次性大负荷运动(负重5 km跑),分别在运动前、运动后0h、3h、12h、24h测定淋巴细胞线粒体DNA(mtDNA)中8-oxodG含量、线粒体膜电位、Bnip3和Beclin-1蛋白表达。结果与运动前比较,运动后0 h和3 h膜电位显著降低,P〈0.01,8-oxodG含量显著升高,P〈0.01,0 h时Bnip3和Beclin-1蛋白表达显著降低,P〈0.05,3 h时Bnip3和Beclin-1蛋白表达显著升高,P〈0.05;运动后12 h与运动后3 h比较,膜电位显著升高,P〈0.05,8-oxodG含量显著降低,P〈0.01,Bnip3和Beclin-1蛋白表达显著升高,P〈0.01;与运动前比较,运动后24 h膜电位、8-oxodG含量、Bnip3和Beclin-1蛋白表达差异无统计学意义,P〈0.05。结论一次性大负荷运动抑制淋巴细胞线粒体自噬,从而造成线粒体功能降低;运动恢复期线粒体自噬活化,清除氧化损伤线粒体,恢复线粒体功能。 Objective To observe the effect of a bout of high-load exercise on mitochondrial autophagy and its biological role in leukocyte. Methods Twenty-four soldiers performed a bout of strenuous weight-loading 5 km running. Leukocyte 8-oxodG content in mitochondrial DNA (mtDNA), mitochondrial membrane potential ( △ ψ), Bnip3 and Beclin-1 protein expression were measured at the moment of pre-training, 0 h, 3 h, 12 h, and 24 h after training respectively. Results As compared with pre-training,△ψdecreased and 8-oxodG content increased in 0 h and 3 h ,P〈0.01. Furthermore, Bnip3 and Beclin-1 expression decreased in 0 h but increased in 3 h ,P〈0.05. As compared with 3 h, △ψ, Bnip3 and Beclin-1 expression increased ,P〈0.05-0.01 but 8-oxodG content decreased in 12 h ,P〈0.01. As compared with pre-training, there was no significant change in △ψ, 8-oxodG content, Bnip3 and Beclin-1 expression in 24 h ,P>0.05. Conclusion A bout of high-load exercise suppressed mitochondrial autophagy and impaired mitochondrial function. During recovery period of exercise, mitochondrial function and oxidative damage were ameliorated via activating mitochondrial autophagy.
