Quality identification of Polygonum hydropiper from Shanxi Province

摘要 为制定陕西产辣蓼药材质量标准及更好地开发利用提供科学依据,采用生药学研究方法、水分测定法、灰分测定法、浸出物测定法、薄层色谱法、挥发油测定法、比色法对陕西产辣蓼药材进行了研究.结果表明:陕西产辣蓼药材水分含量7.42%~8.61%,总灰分含量8.06%~8.65%,酸不溶性灰分含量0.23%~0.74%,醇溶性浸出物含量19.86%~23.62%,挥发油含量0.13~0.17 mL/g,总黄酮含量2.01%~4.59%.其中,洋县、柞水县辣蓼的醇溶性浸出物较高,略县、柞水县辣蓼的黄酮含量较高,蓝田县辣蓼的醇溶性浸出物和黄酮含量都最低,其他地区的辣蓼介于中间.陕西柞水辣蓼药材品质最好,而蓝田辣蓼的品质最差;建立了以性状描述、显微特征描述、指标有效成分芦丁的薄层定性鉴别及水分含量不得过于9%、总灰分含量不得过于9%、酸不溶性灰分不得过于1%、醇溶性浸出物不得少于15%、总黄酮含量不得少于2%为主要内容的陕西辣蓼质量标准. To provide scientific basis for the development and utilization of P.hydropiper from Shanxi, TLC,essential oil and UV-vis spectra determination methods were employed to analyze moisture,total ash ,acid-insoluble ash,content of the extraction.The results showed that moisture content was 7.42% to 8.61%,total ash content was 8.06% to 8.65%,acid-insoluble ash content was 0.23% to 0.74%,content of ethanol extraction was 19.86% to 23.62%,essential oil content was 0.13% to 0.17% and total fla-vonoids content was 2.01% to 4.59%.The content of ethanol extraction from Yang County and Zhashui County was higher than the other production areas.Total flavonoids content from Lue County and Zhashui County was higher.The content of ethanol extraction and total flavonoids from Lantian County showed the lowest.It is suggested that the quality of P.hydropiper from Zhashui County is best and the one from Lantian County is the worst.The quality identification of P.hydropiper from Shanxi was finally confirmed as follow:the morphological and histological characters were described.Rutin content was chosen as the mark constituent in qualitative identification analysis of the crude drug by TLC method.Moisture was be-low 9%,total ash was below 9%,acid-insoluble ash was below 1%,content of ethanol extraction was above 1 5% and total flavonoids was above 2%.
