摘要
目的:以灵芝多糖、红景天苷和西洋参皂苷为例,探讨不同剂量和给药时程联合用药的抗肿瘤作用及对荷瘤小鼠细胞免疫功能的影响。方法:灵芝多糖、红景天苷和西洋参皂苷按10∶10∶1.5比例组合,以SPF级昆明小鼠为实验对象,随机分为正常对照组(A)、模型组(B)、低剂量持续给药组(C)、低剂量预防给药组(D)、高剂量持续给药组(E)和高剂量预防给药组(F),C^F组灌胃相应剂量的受试组合物,A组和B组灌胃等量生理盐水。连续灌胃4周后,除正常对照组外其余各组小鼠均接种肉瘤S180细胞,持续给药组继续灌胃。接种3周后处死小鼠,检测各组荷瘤小鼠肿瘤大小、细胞免疫功能及血清中IL-2、IFN-γ和TNF-α含量。结果:受试组合物低剂量持续给药和低剂量预防给药均能显著抑制S180实体瘤的生长(P<0.05),并明显延长荷瘤小鼠生存期(P<0.05),均能显著提高小鼠细胞免疫功能以及血清IL-2、IFN-γ和TNF-α水平,以低剂量预防给药效果最为显著。高剂量预防给药可以抑制S180实体瘤的生长,延长荷瘤小鼠生存期,并能提高细胞免疫功能及血清IL-2、IFN-γ和TNF-α水平,但与模型组相比无显著性差异(P>0.05)。然而高剂量持续给药却能抑制ConA诱导的小鼠淋巴细胞转化能力,降低荷瘤小鼠的血清IL-2、IFN-γ和TNF-α水平,显著缩短荷瘤小鼠生存期(P<0.05),具有促进肿瘤生长的趋势。结论:受试组合物在合适剂量和给药时程下可增强荷瘤小鼠的细胞免疫功能,延长荷瘤小鼠生存期,抑制肿瘤细胞生长,但是剂量过大且持续给药会抑制细胞免疫功能,缩短生存期,有促进肿瘤生长的倾向。
Objective: Using combination of Ganoderma lucidum polysaccharides,Salidroside and Panax quinquef olium saponin as an example,to investigate effects of different doses and administration schedules on suppressing tumor growth and modulating the cellular immunity in mice bearing tumors derived from sarcoma S180 cells. Methods: Kunming mice(SPF degree) were selected,and mice were randomly divided into six groups: the control group( A),model group(B),low-dose consistent administration group(C),low-dose prophylactic administration group(D),high-dose consistent administration group(E) and high- dose prophylactic administration group(F). Mice of C ~ F groups were given the tested combination(10:10:1. 5) by intragastric administration,and mice of A ~ B groups were given physiological saline. After corresponding treatments for 4 weeks,all mice were inoculated subcutaneously with S180 cells except mice in group A. Mice in group C and group E were still subjected to treatment. After inoculation for 3 weeks,the tumor inhibitory rate was calculated,the cellular immunity function,IL- 2,IFN- γ and TNF- α levels of the mice were detected. Results:The low- dose combination significantly suppressed the tumor growth,improved cellular immune function,and increased the levels of serum IL- 2,IFN- γ and TNF- α(P〈0. 05),and the effect of low- dose prophylactic administration was particularly significant(P〈0. 01). The high- dose prophylactic administration also suppressed the tumor growth and improved cellular immunity,but had no significant difference from the model group(P〈0. 05). However,high- dose consistent administration decreased the ability of ConA induced lymphocytes transformation,decreased the levels of serum IL- 2,IFN- γ and TNF- α,significantly shortened survival of tumor- bearing mice( P〈0. 05),and tended to promote tumor growth. Conclusion: A suitable dose and administration schedule of the tested combination can suppress the growth of S180 implant tumor and enhance the cellular immunity of the tumor- bearing mice. High dosage consistent administration can suppress cell- mediated immune function,shorten survival of tumor- bearing mice,and tend to promote tumor growth.
