气道滴入脂多糖前后用全氟化碳对小鼠急性肺损伤的影响 被引量:2

摘要 目的探讨气道滴入脂多糖前后全氟化碳(PFC)对小鼠急性肺损伤(ALI)的影响。方法雄性ICR小鼠52只,分为2批,第1批32只,第2批20只,每批分为4组,正常对照组:0.9%氯化钠注射液(NS)对照组;脂多糖(LPS)组:采用LPS气道滴入诱导小鼠ALI模型,P1组:LPS滴入30min前用PFC,P2组:LPS滴入30min后用PFC。LPS气道滴入后3h处死动物,第1批左肺灌洗,检测白细胞及中性粒细胞计数,右肺上叶石蜡切片苏木素-伊红(HE)染色,做病理分析,中下叶酶联免疫吸附试验(ELISA)法检测肺组织匀浆肿瘤坏死因子(TNF)-α、白细胞介素(IL)-8、肺组织基质金属蛋白酶2(MMP-2)含量;第2批处死前30min尾静脉注射1%伊文斯蓝染料0.1mL,取左肺测定湿质量/干质量比、取右肺测肺通透性。结果 PFC降低肺湿质量/干质量比及肺通透性,降低肺泡灌洗液中白细胞总数和中性粒细胞数比例,抑制组织MMP-2的释放,降低肺组织匀浆TNF-α、IL-8的含量,LPS滴入前比滴入后使用PFC对上述各指标的作用更明显(P<0.05)。结论气道滴入LPS前比滴入后用PFC更能减轻LPS诱导的ALI,可能通过减少肺组织产生TNF-α减轻肺损伤。 Objective To investigate the effects of perfluorocarbon(PFC)before and after intratracheal LPS administration on acute lung injury.Methods Fifty-two male ICR mice were prepared.Thirty-two mice were randomly divided into 4groups(n=8),C group:normal saline(NS)group,L group:LPS group,P1group:PFC at 30 min before LPS instillation,P2group:PFC at 30 min after LPS instillation.The animals were executed at 3hours after LPS instillation.The lung wet/dry weight ratio,lung permeability were examined.Another 20 mice were also divided into the same 4groups(n=5).Matrix metalloproteinase-2(MMP-2),TNF-α,IL-8content in lung tissue were examined by ELISA.Neutrophils in bronchoalveolar lavage fluid(BALF)were detected.Results PFC reduced the lung wet/dry weight ratio,reduced the total number of white blood cells and neutrophil number proportion in BALF,inhibited MMP-2release,reduced the content of TNF-αand IL-8in lung tissue.Using PFC before LPS instillation have more obvious effects on each index than after(P〈0.05).Conclusion Using PFC before LPS instillation have better protective effects on ALI by reducing TNF-αin lung tissue.
