黄芪注射液对大鼠皮层神经细胞缺氧性凋亡的影响

摘要 目的观察黄芪注射液对大鼠皮层神经细胞缺氧性凋亡的影响。方法体外原代培养20只SD大鼠脑皮质神经细胞,种入96孔板,分为对照组、模型组和黄芪组。黄芪组与模型组制作缺氧性凋亡细胞模型。黄芪组分别加入1、10、100μg/mL的黄芪注射液,模型组及对照组不用药。干预后12、24 h采用流式细胞仪分析细胞凋亡情况;AO/EB双重染色、透射电子显微镜观察细胞形态结构变化;Western blot法检测Bcl-2、Caspase-8蛋白表达。结果荧光显微镜及电镜下,黄芪组神经细胞形态学改变较模型组轻,细胞凋亡数量较少。黄芪组、模型组细胞凋亡率高于对照组(P均<0.05),但黄芪组神经细胞凋亡率低于模型组,且凋亡率随黄芪浓度增加而降低(P均<0.05)。黄芪组、模型组Bcl-2表达均低于对照组,但黄芪组高于模型组,且Bcl-2表达随黄芪浓度增加而增强(P均<0.05)。黄芪组、模型组Caspase-8表达高于对照组,但黄芪组低于模型组,且Caspase-8表达随黄芪浓度增加而降低(P均<0.05)。结论黄芪注射液可抑制大鼠皮层神经细胞缺氧性凋亡,其机制可能与调控Bcl-2、Caspase-8蛋白表达有关。
