摘要 以鸡传染性支气管炎病毒(Infectious Bronchitis Virus,IBV)的N基因保守区域设计4条引物,通过优化反应条件,建立了检测IBV的逆转录环介导等温扩增技术(RT-LAMP),并进行了特异性试验、敏感性试验及临床样本检测。结果显示,建立的RT-LAMP方法对IBV参考株扩增产物的电泳呈特征性梯状条带,而新城疫病毒(NDV)、禽流感病毒(AIV)、马立克病病毒(MDV)、传染性囊病病毒(IBDV)等的扩增产物电泳后均无扩增条带。该方法最低能检出0.19 pg的IBV cDNA模板,敏感性比常规RT-PCR方法高1 000倍。用该方法检测了6份临床疑似IB临床病料,检出率为100%(6/6),与常规RT-PCR的符合率为100%。60 min反应结束后,离心可以看到白色沉淀物,简单直观,适用于基层实验室快速准确诊断。 Designed 4 primers of the N gene of conservative region for IBV, the specificity test, sensitivity test and the detection of clinical samples were experimented too. The results showed that using the RT-LAMP,the amplified products of reference strain for IBV in electrophoresis presented characteristic bands, which were not found in NDV、AIV 、MDV and IBDV. The minimum detectable amount of cDNA of IBV was 0.19 pg by using this RT-LAMP method, and the sensitivity was 1000 times higher than conventional RT-PCR method. The detection rate of six clinical samples was 100% which was highly consistent with the result by using conventional RT-PCR. 60 min after the end of the reaction,centrifuged the reaction product, white precipitate could be seen. The method was simple and intuitive and suitable for fast and accurate diagnosis of grassroots laboratory detection.
