摘要
用活性炭吸附糖化酶,再以不同配比的聚乙烯醇-海藻酸钠复合溶胶覆膜,分别对固定化条件、米氏常数、酶活残留率进行了研究,结果表明:活性炭覆海藻酸钠复合膜固定化方法为最佳,即海藻酸钠:聚乙烯醇配比为8:2,溶胶浓度为1.5%,固化时间为120min。该方法得到的固定化酶米氏常数为3.6mg/mL,重复使用七次后其酶活残留率为88%。
With activated carbon adsorption immobilized glucoamylase, with different ratio of polyvinyl alcohol (PVA) and sodium alginate composite sol, laminating of conditions of immobilized, Michaels constant and enzyme activity retention rate were studied, the results show that the best immobilized method was activated carbon by sodium alginate composite membrane: the ratio of sodium alginate : PVA for 8□ 2, sol concentration as 1.5%, the curing time of 120 rain. Miehaelis constant as 3.6 mg/mL, after repeated use seven times its enzyme activity retention rate of 88%.enzyme activity retention rate as 88%.
