摘要
目的 研究人工关节材料磨屑诱发骨溶解的机制。 方法 超高相对分子质量聚乙烯 (ultrahighmolecularweightpolyethylene ,UHMWPE)磨屑在体外与Ana- 1小鼠巨噬细胞 (macroph age,M)共同培养 ,测定白细胞介素 - 1β(IL - 1β)、肿瘤坏死因子 -α(TNF -α)的含量。 结果 磨屑数量 >10 6 pg ml才能引起明显的生物学反应 ,≤ 10 μm的磨屑引起的生物学反应与对照组比较 ,差异有非常显著性意义 (P <0 .0 1)。 结论 M释放骨溶解因子的能力与磨屑大小、数量有关 ,减少磨损、降低假体周围组织中的磨屑量 。
Objective To study the mechanism of osteolysis induced by prosthesis material polyethylene particles. Methods In vitro Ana 1 macrophages were cultured and stimulated by ultrahigh molecular weight polyethylene (UHMWPE) particles with different concentration or size. The levels of TNF α and IL 1β of the supernatants were measured by radioimmunoassay. Results Macrophages exposed to wear particles released TNF α and IL 1β in dose and size dependent fashions. Only over 10 6 pg/ml of particles could induce bioeffects. The levels of TNF α and IL 1β were significantly increased when the macrophages were exposed to less than 10 μm of particles. Conclusions Elimination of wear particles is a principal method to prevent aseptic loosening of prostheses.
