摘要
目的观察普罗布考对非酒精性脂肪性肝病(NAFLD)大鼠线粒体的影响。方法 24只雄性SD大鼠随机分为3组:正常对照组(NC组)、NAFLD对照组(HF组)、普罗布考干预组(PB组)。以高脂饲料饲养诱发脂肪肝大鼠模型。18周末,处死所有动物,检测血脂、肝功能、大鼠肝线粒体丙二醛(malondaldehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、肝线粒体膜电位、肝组织匀浆ATP酶等。结果与NC组比较,HF组TC、LDLC、AST、ALT、肝线粒体MDA含量、肝线粒体膜电位显著升高,肝线粒体SOD和GSH-Px活性、Na-K ATP酶、Ca-Mg ATP酶、HDLC显著降低,差异均有统计学意义(P<0.05);与HF组比较,PB组TC、LDL-C、AST、ALT、肝线粒体MDA含量、肝线粒体膜电位显著下降,肝线粒体SOD、GSH-Px活性、Na-K ATP酶、Ca-Mg ATP酶显著升高,差异均有统计学意义(P<0.05),但HDL-C差异不显著(P>0.05);与NC组比较,PB组HDL-C显著降低(P<0.05)。结论普罗布考可减轻NAFLD大鼠肝脏线粒体氧化应激的损害。
Objective To investigate the effects of Probucol on mitochondria of nonaloholic fatty liver disease (NAFLD) in rats.Methods Twenty-four male SD rats were randomly divided into normal control group (NC group),NAFLD control group (HF group) and Probucol prevention group (PB group).NAFLD rat model was established by feeding with high caloric diet.At the end of the 18th week,all the rats were sacrificed to dectect serum TC,TG,ALT,AST and the changes of MDA,SOD and GSH-Px of liver mitochondria,MMP and liver hemogenate ATP enzyme and so on.Results Compared with NC group,serum TC,ALT,LDL-C,AST,MDA and MMP of liver mitochondria were increased significantly; SOD,GSH-Px,Na-K ATP,Ca-Mg ATP,HDL-C were decreased markedly in HF group (P <0.05).Compared with HF group,serum TC,ALT,LDL-C,AST,MDA and MMP of liver mitochondria were reduced markedly,SOD,GSH-Px,Na-K ATP,Ca-Mg ATP were increased significantly in PB group (P <0.05),while except HDL-C.Compared with NC group,HDL-C was decreased significantly in PB group.Conclusion Probucol reduce the liver oxidation stress on mitochondria of NAFLD in rats.
