摘要
目的:研究脉络宁注射液对脂多糖与角叉菜胶诱导"热毒血瘀证"模型大鼠的核转录因子κB(NF-κB)和肿瘤坏死因子α(TNF-α)的作用。方法:50只SPF级SD雄性大鼠随机分为空白对照组,模型对照组和脉络宁高、中、低剂量组(30mg/kg,15mg/kg,3.75mg/kg),每组10只,腹腔注射给药20天,每天1次。给药到18天,除空白对照组外,其余各组大鼠腹腔注射Ca(50mg/kg)16h后,再尾静脉注射LPS(5μg/kg),24h后大鼠尾部出现瘀血现象为模型建立成功。给药20天后,从腹主动脉取血和取脾。检测血液的红细胞比积和活化部分凝血活酶时间,光镜下观察脾脏的病理变化,免疫组化法检测各组大鼠脾脏的核转录因子κB(NF-κB)和肿瘤坏死因子α(TNF-α)。结果:与模型对照组比较,脉络宁高剂量组的红细胞比积,NF-κB和TNF-α显著减少(P<0.05),活化部分凝血活酶时间显著缩短(P<0.05),脾脏的脏器系数显著增加(P<0.05)。结论:脉络宁注射液对脂多糖与角叉菜胶诱导"热毒血瘀证"模型大鼠有一定的活血抗炎作用。
Objective: To study the effect ot Mattuonmg lnjecuon( TNF)on NF -κB, els with noxious heat stasis syndrome induced by lipopolysaceharide(LPS) combined with carrageenan (Ca). Methods: Fifty SPF SD male rats were randomly divided into a blank control group, a model control group and high, middle and low dose of Mailuoning groups (30mg/kg, 15mg/kg,3.75mg/kg). Rats in the model group were pretreated with Ca i.p. at the doses of 50mg/kg, then treated with LPS i.v. at the doses of 5 μg/kg six- teen hours later. After 24 hours, rat tails had the phenomenon of congestion which was considered as success-ful rat models. Blank control and model control groups were injected with the same capacity of NS. Low, mid- dle and high dose of Mailuoning groups were given Mailuoning by injecting intraperitoneally (3.75mg/kg, 15mg/kg,30mg/kg) , for 20 consecutive days, once a day. After the first 18 days administration, rats of mod- el control group and each dose of Mailuoning groups were being models. Their blood was collected from the ab- dominal aorta and removed spleen. We detected red blood cells and measured the activated clotting time and observed the histopathology of spleen by light microscopy. Results: Red blood cells and the expressions of NF - κB and TNF - α correspondingly were reduced ( P 〈 O. 05 ) ; the activated clotting time was shortened (P 〈 O. 05 ), and the weight of spleen increased ( P 〈 0.05 ). There were no obvious inflammation and con- gestion symptoms in the high dose of Mailuoning group compared with those of model group. Conclusion : Mai- luoning injection has an anti - inflammatory effect on the rat models with noxious heat stasis syndrome induced by lipopolysaccharide combined with carrageenan.
