摘要
选取经驯化栽培的山樱花植株茎尖和带腋芽的半木质化茎段为外植体,对其组培快繁体系进行研究的结果表明:山樱花茎段外植体在MS+6-BA 0.6mg/L+NAA 0.02mg/L+Vc 40mg/L+3%Sucrose+0.7%Agar(pH5.8)培养基上,腋芽萌发率为90.0%以上;其组培苗在MS+6-BA 0.4mg/L+NAA 0.02mg/L+Vc 40mg/L+3%Sucrose+0.7%Agar(pH 5.8)培养基上继代培养25天左右,继代增殖系数可高达4.0以上;将长势良好的组培苗在1/2MS+NAA 0.8mg/L+Vc 40mg/L+1.5%Sucrose+0.7%Agar(pH 5.8)培养基上生根壮苗培养后,移栽成活率可达73.3%。
The studied of rapid propagation of domesticated Cerasus serrulata by tissue culture with stem apex or stem sections with axillary buds as explants. The result showed that more than 90% of axillary buds germinated on the primary medium contained MS basal salts and vitamins, 6 -BA0. 6rag/L, NAA0. 02mg/L, Vc 40mg/L and 3% sucrose which was solidified with 0. 7% agar(pH 5.8). After the shoots in vitro were subcultured for about 25 days, the subculture medium contained MS basal salts and vitamins, 6 - BA0. 4rag/L, NAA0. 02mg/L, Vc 40mg/L and 3% sucrose which was solidified with 0.7% agar(pH 5.8), the propagation coefficient by subculture was more than 4. 0. The shoots were rooted and grew stronger on thel/2 MS medium contained NAA0. 8 mg/L, Vc 40mg/L and 1.5% sucrose, and the transplanted survival rate is 73.3%.
