摘要
目的:观察舒心稳斑颗粒对实验兔AS斑块稳定性及MMP-9、I型胶原蛋白表达的影响。方法:运用高脂高胆固醇饲料喂养12周制备日本大耳白兔动脉粥样硬化模型,用舒心稳斑颗粒灌胃给药治疗4周,采用双抗体夹心酶联免疫吸附法检测血清中I型胶原蛋白的含量;采用免疫组化法分析I型胶原蛋白、MMP-9在主动脉粥样斑块中的表达;同时观察主动脉粥样硬化斑块的病理学改变。结果:与模型组比较,舒心稳斑颗粒能升高血清中Ⅰ型胶原蛋白的含量,差异具有显著统计学意义(P<0.01);同时能降低斑块中MMP-9的表达水平,升高Ⅰ型胶原蛋白在斑块中的表达水平。病理学检查结果显示,采用舒心稳斑颗粒干预后,主动脉粥样硬化斑块的病理学形态、结构均发生改变,斑块结构更加趋于稳定。结论:舒心稳斑颗粒可能通过抑制斑块内炎症反应,调节动脉粥样硬化斑块纤维帽中的胶原纤维降解与合成失衡状态,进而对AS斑块起稳定作用。
Objective:To observe the effects of Shuxin wenban granule on the stability of AS plaque and the expressions of MMP-9 and COL-l in rabbits.Methods: Using the high cholesterol diet to induce AS rabbit models for 12 weeks, then, giving Shuxin wenban granule for 4 weeks, finally,detecting the serum levels of COL-I in the rabbits by double antibody sandwich enzyme -linked immunosorbent method and the surface density of MMP-9 and COL-I in atherosclerotic plaques by immunohistochemical staining .At the same time , observing the pathological changes of aorta in atherosclerotic rabbits .Results:To compared with model group , Shuxin wen-ban granule could increase the serum levels of COL -I,the difference was statistically significant (P〈0.01);similarly, Shuxin wenban granule could significantly decrease the surface density of MMP -9 and increase the surface density of COL-I in atherosclerotic plaques,the difference was statistically significant (P〈0.01).Pa-thology showed that Shuxin wenban granule changed the pathological morphology , structure of atherosclerotic plaque , caused the plaque to stabler .Conclusion: Shuxin wenban granules could stabilize atherosclerotic plaques by inhibiting the inflammatory response of atherosclerotic plaque and affecting the imbalance of collagen fibers’ degradation and synthesis in atherosclerotic plaque fibrous cap .
