摘要
目的:观察姜黄素对海人酸活化的BV-2小胶质细胞增殖的影响,探讨其可能的分子机制。方法:将BV-2细胞分为正常对照组、海人酸组和姜黄素组,倒置显微镜观察姜黄素对海人酸活化的BV-2细胞形态的影响,MTT法检测姜黄素对海人酸活化的BV-2细胞增殖的影响,免疫组化法检测姜黄素对海人酸活化的BV-2细胞PKC-δ蛋白表达的影响。结果:倒置显微镜观察结果显示,海人酸组与正常对照组相比BV-2细胞数量增加、胞体变大、突起变短变粗;姜黄素组与海人酸组相比BV-2细胞数量减少、胞体变小、突起变长变细。MTT检测结果显示,5μmol/L姜黄素作用BV-2细胞2-96小时,BV-2细胞抑制率随着姜黄素作用时间的增加逐渐增高;5μmol/L姜黄素作用BV-2细胞2-8小时,实验组BV-2细胞抑制率与海人酸组相比无显著差异(P〉0.05),5μmol/L姜黄素作用BV-2细胞16-96小时,姜黄素组BV-2细胞抑制率与海人酸组相比具有显著差异(P〈0.05)。免疫组化检测结果显示,海人酸组BV-2细胞PKC-δ表达明显高于对照组(P〈0.05);与海人酸组相比,姜黄素组BV-2细胞PKC-δ表达明显减弱(P〈0.05)。结论:姜黄素可抑制海人酸活化的BV-2细胞增殖,其机制可能与下调BV-2细胞PKC-δ蛋白表达有关。
Objective: To observe the effects of curcumin on the proliferation and expression of PKC - protein in kainite- activated BV -2 cells and investigate the related molecular mechanism. Methods: BV -2 cells were randomly divided into three group: control group, kainite group and earcumin group. The effect of curcumin on the morphology of kainate - activated BV - 2 cells was observed with inverted microscope, the effect of curcumin on the proliferation of kainate - activated BV - 2 cells was detected with MTT assay, the effect of curcumin on the expression of PKC - 8 protein in kainate - activated BV - 2 cells was determined by imunohisto- chemistry. Results: The inverted microscope showed that the number of BV - 2 cells in kainite group increased, the volume get larger and protuberance become shorter and coarser to compare with control group, the number of BV - 2 ceils in eurcumin group decreased, the volume get smaller and protuberance become longer and thinner to compare with the kainite group. MTT assay result showed that the inhibition rates of 5μmol/L cureumin for BV - 2 cells between 2h and 96h increased gradually with the increasing of time, there was no obvious difference in the inhibition rates of 5 μmol/L curcumin between 2h and 8h to compare with the kainite group ( P 〉 0. 05 ), there was obvious difference in the inhibition rates of 5μmol/L eurcumin between 16h and 96h to compare with kainite group (P 〈 0. 05). Immunohistochemistry technique resut showed that the expression of PKC - 8 protein in BV - 2 cells of kainite group was higher than that of control group(P 〈0. 05) ; the expression of PKC -8 protein in BV - 2 cells of cureumin group was lower than that of kainate group(P 〈 0. 05). Conclusion: Curcumin can in- hibit the proliferation of BV - 2 cells and the mechanism may be associate with down - regulating the expression of PKC - δ protein.
