麝香酮联合灯盏花素对TBI大鼠脑血流量及caspase-3蛋白表达的影响 被引量:6

Effects of muscone combined breviscarpin on cerebral blood flow and caspase-3level in rats with traumatic brain injury

摘要 目的探讨麝香酮联合灯盏花素治疗创伤性颅脑损伤(TBI)大鼠的治疗效果,并与单用灯盏花素进行比较。方法选择SD大鼠50只,随机分为假手术组、TBI模型组、麝香酮组(1 mg·kg-1)、灯盏花素(200 mg·kg-1)组和联用组,均n=10。采用液压打击法制造大鼠左侧TBI模型,假手术组只开骨窗。分别于制模前6 h、30 min,制模后30 min、6 h给予相应剂量的灯盏花素或麝香酮灌胃给药。激光多普勒血流仪检测各组制模前30 min及制模后30 min、6 h、12 h和24 h的额叶大脑皮质血流量(CBF)变化。制模后24 h,检测大鼠学习记忆能力、脑含水量及caspase-3蛋白表达的情况。结果与模型组比较,麝香酮组各指标无显著差异(P>0.05),灯盏花素组在制模后12 h、24 h CBF显著提高(P<0.05),联用组在各时间点CBF均显著高于模型组(P<0.01)。联用组较模型组和灯盏花素组学习记忆能力均显著提高(P<0.05或P<0.01),脑含水量显著降低(P<0.01),且caspase-3阳性细胞的表达显著降低(P<0.01)。结论麝香酮联合灯盏花素治疗急性重型TBI比单用灯盏花素具有更好的治疗作用。 AIM To investigate the therapy effect of muscone combined breviscarpin in rats with traumatic brain injury (TBI), comparing with breviscarpin. METHODS A total of 50 SD rats were randomly divided into sham group, TBI model group, nmscone group ( 1 mg·kg^-1) , breviscarpin (200 mg·kg^-1) group and muscone (1 mg·kg^-1) combined breviscarpin (200 mg·kg^-1) group, n = 10 in each group. TBI models were made in left brain of rats by hydraulic shock, while rats were only opened bone window in the sham group. Rats in muscone group, breviscarpin group and muscone combined breviscarpin group were administrated by ig at 30 min and 6 h before TBI and 30 min and 6 h after TBI. The change of cerebral blood flow (CBF) at cortex of frontal lobe was detected at the time 30 min before TBI and 30 min, 6 h, 12 h and 24 h after TBI by laser doppler flowmetry. The rats' learning and memorizing ability, contents of brain water and caspase-3 level were detected at the time 24 h after TBI. RESULTS Compared with the model group, there were no significant difference in muscone group (P 〉 0.05). CBF of the breviscarpin group was increased significantly at 12 h and 24 h after TBI (P 〈 0.05). CBF of the muscone combined breviscarpin group was higher than model group at each time after TBI (P 〈 0.05 or P 〈 0.01 ). The contents of brain water of muscone combined breviscarpin group was decreased significantly (P 〈 0.01). The caspase-3 level of muscone combined breviscarpin group was decreased significantly (P 〈 0.01). CONCLUSION In TBI therapy, better treatment effects will be obtained when muscone combined breviscarpin than breviscarpin alone.
