摘要
目的探讨microRNA-23b与MG发病的相关性。方法 31例MG患者,31例健康人,取其外周血分离外周血单个核细胞(PBMCs),提取RNA并逆转录,实时定量PCR检测microRNA-23b的表达,以小分子RNAU6为内参,计算标准化后的2-ΔCt值表示microRNA-23b相对表达量。结果 MG患者PBMCs中microRNA-23b的表达水平高于健康对照组(P=0.0266),但其在MG不同临床特征亚组中的表达差异无统计学意义(P>0.05)。MicroRNA-23b表达水平与肌无力严重程度之间的相关性无统计学意义(r=-0.0308,P=0.8695)。结论 MG患者PBMCs中microRNA-23b异常高表达,但与性别、发病年龄、类型、胸腺状态、甲状腺功能及肌无力严重程度无关。
Objective To investigate the expression of microRNA-23 b in peripheral blood mononuclear cells( PBMCs) of patients with myasthenia gravis( MG). Methods PBMCs of 31 patients with MG were separated from the peripheral blood( the study group) and 31 healthy individuals( the control). Then total RNAs were isolated,the levels of microRNA-23 b were determined by quantitative reverse transcription PCR( qRT-PCR),using U6 as housakeeping control. Relative expression levels were expressed as 2- ΔCt. Results The expression of microRNA-23 b in PBMCs was significantly higher in MG group than in control group( P = 0. 0266). The expression of microRNA-23 b in MG patients with different clinical characteristics had no statistic difference( P 〉0. 05). The correlation between the expression level of microRNA-23 b and muscle weakness severity had also no statistic difference( r =- 0. 0308,P = 0. 8695). Conclusions The expression of PBMCs microRNA-23 b was higher in MG patients. However,no statistically significant relationships were found between microRNA-23 b expression levels and demographic or clinical characteristics in the Patients with MG.
