摘要
目的本文通过研究染料木素(Gen)对体外培养MGC-803细胞增殖以及PKA活性的影响,探讨Gen抑制MGC-803细胞系增殖的分子机制。方法光镜下观察经Gen处理后人胃癌MGC-803细胞形态学改变;应用MTT法检测Gen对体外培养的人胃癌MGC-803细胞增殖影响;Pep Tag法检测Gen对MGC-803细胞PKA活性的调节。结果与空白对照组比较,经Gen处理后MGC-803细胞外形呈不规则状、细胞间隙增大、折光性差;MTT实验发现Gen能够抑制体外培养的MGC-803细胞增殖,并且抑制作用呈现时间及浓度依赖性;浓度为40μg/m L的Gen可以显著上调MGC-803细胞的PKA活性(P<0.01)。结论 Gen对人胃癌细胞系MGC-803细胞的增殖有抑制作用,上调PKA活性可能是Gen抑制人胃癌MGC-803细胞增殖的机制之一。
Objective To study the effects of Genistein on proliferation and the activity of PKA of MGC-803,In order to find potential molecular mechanism of anti-MGC-803 cell lines of Gen. Methods Cell morphology was observed by inverting microscope. The inhibitory of Gen was determined by MTT assay on human gastric cancer cell lines of MGC - 803 in vitro; The activity of PKA was detected by PepTag Non-Radioactive PKA Assay. Results The morphology of Gen group was irregular cell shape, cell gap increased, refractive index of cell turn into weak and poor growth state. Compared with the blank control group,the activity of PKA was increased of statistic significance in the Gen group (P〈 0.05). Conclusion The proliferation of human gastric carcinoma MGC-803 cells can be inhibited by Gen, the up-regulation of PKA activity may be one of mechanisms of anti-MGC-803 cell lines of Gen.
