S1P在臀肌挛缩症发病过程中的表达及作用 被引量:1

摘要 目的探讨磷酸鞘氨醇(sphingosine-1-phosphate,S1P)在臀肌挛缩带中的表达情况及其对臀肌挛缩症(gluteal muscle contracture,GMC)发病的作用。方法收集臀肌挛缩症病人病理标本,采用免疫组织化学、Western blot和酶联免疫吸附技术(ELISA)观察S1P在臀肌挛缩带中的表达特征,以及其对瘢痕成纤维细胞增殖及Ⅰ型胶原蛋白分泌的作用。结果S1P定位于成纤维细胞的胞质中,呈强阳性表达,瘢痕成纤维细胞在S1P作用下呈时间及浓度依赖性增殖,且最高增殖达3.2倍(P<0.05)。Ⅰ型胶原和α-平滑肌动蛋白(α-SMA)也呈现出对S1P浓度依赖性表达,其最高表达分别为初始值的1.4倍和3倍(P<0.05)。结论 S1P的高表达及其对瘢痕成纤维细胞增殖及Ⅰ型胶原和α-SMA的正性调控可能是引起臀肌挛缩症发病的重要机制。 Objective To explore the expression and role of sphingosine-1-phosphate (S1P) in gluteal muscle contracture (GMC).Methods The pathological specimens from GMC patients were collected to investigate the expression of S1P and its effect on cell proliferation,collagen secretion and myofibroblast transformation by immunohistochemistry,ELISA and Western bolt.Results S1P was intensely expressed in fibroblast cells.S1P increased scar-derived fibroblasts proliferation in a time-and dose-dependent manner up to the 3.2-fold(P < 0.05).S1P also promoted collagen I production and α-SMA expression,which was up to 1.4-fold and 3-fold,respectively(P < 0.05).Conclusion Increased expression of S1P and its effect on fibroblasts proliferation,collagen I production and α-SMA expression may be an important mechanism in the process of gluteal muscle contraction.
