摘要
NOTCH1信号通路是影响机体发育的重要通路,而研究NOTCH1的靶基因及其功能对阐明该通路的分子机制非常有意义。NOTCH1蛋白的入核片段ICN1,通过招募共转录因子CSL和MAML1形成转录复合物,启动靶基因的表达。通过生物信息学分析预测了21个潜在的NOTCH1靶基因。半定量PCR、定量PCR和蛋白表达分析的结果表明SEPT4是NOTCH1的一个新的靶基因。
NOTCH1 signaling pathway is very important for body development and the study of functions of NOTCH1 target genes is helpful to understand molecular mechanisms. The activated fragment of NOTCH1,ICN1,enters the cell nucleus and recruits its partners( co-activators) : CSL and MAML1,to form a DNA-binding complex and initiates the transcription of NOTCH1 target genes. Bioinformatics predicts that there are 21 new possible NOTCH1 target genes. The results of semi-quantitative PCR,real-time quantitative PCR,and protein expression analyses indicate that SEPT4 gene is a NOTCH1 target gene.
