间充质干细胞的免疫学特点及其应用

摘要 目的:探讨间充质干细胞的免疫学特点及其在医学上的应用。方法:肝脏移植术后免疫排斥反应较严重,患者一般须严格服用免疫抑制剂。本实验以肝脏移植为例,研究MSC的免疫学特点。首先分离纯化大鼠骨髓间充质干细胞及普通造血干细胞,并建立两组大鼠肝脏移植后急性排斥反应的模型,分为实验组和对照组,实验组给予间充质干细胞培养的肝脏组织移植,对照组接受普通干细胞培养的肝脏组织移植。两组模型均给以半剂量的免疫抑制剂,观察实验组的MSC能否减轻其免疫反应,并与未加入MSC的对照组作对比。结果:实验组仍无明显免疫排斥反应,术后1周饮食及精神状况良好,生存质量与正常健康大鼠无明显差异。表明增强机体的免疫耐受,减弱免疫排斥反应。对照组除总胆红素含量外,其他各项指标均有不同程度偏高。实验组存活率高达91.7%,而对照组仅33.3%。结论:间充质干细胞具有低免疫原性,并可诱导机体免疫耐受能力,临床上可在器官移植等疾病上推广应用。 Objective:To investigate the immunological characteristics and applications of mesenchymal stem cells.Methods:Rejection reaction after liver allograft is more serious,patients generally should be strictly taken immunosuppressants.In this experiment,a liver transplant was taken for example,the immunological characteristics of the MSC was studied.First,mesenchymal stem cells and ordinary hematopoietic stem cells in rat bone marrow was separated and purified,and acute rejection reaction models were established after liver transplantation in two groups,divided into experimental group and control group.Two models were given a half dose of immunosuppressant,the immune response in the experimental group with MSC was observed,and with the control group without MSC for comparison.Results:The experimental group was still no significant immune rejection,after a week eating and spirit was good,the quality of life had no significant difference compared with normal healthy rats,it showed immune tolerance was enhanced and immune rejection was weakened.In the control group,except total bilirubin levels,other indicators were higher in varying degrees.Survival rates in the experimental group was as high as 91.7%,while the control group,only 33.3%.Conclusion:Mesenchymal stem cells have low immunogenicity and induce immune tolerance,can promote the use of organ transplantation in clinical.
