摘要
目的研究切除精索神经对Leydig细胞凋亡和PARP、Fas基因表达的影响。方法成年健康SD大鼠20只,随机分为假手术组和脊髓损伤组,手术后14 d以Percoll连续密度梯度法提取Leydig细胞后采用流式细胞仪(FACS)定量检测Leydig细胞凋亡情况,以Western blot方法检测Leydig细胞中cleaved-PARP、Fas蛋白的表达。结果脊髓损伤组凋亡Leydig细胞比例(32.15%)显著高于假手术组(12.78%)(P<0.05),其Fas蛋白及Cleaved-PARP蛋白的表达水平均显著升高。结论脊髓损伤诱发Leydig细胞凋亡,PARP、Fas基因参与对该过程的调控。
Objective To explore influence of spinal cord injury on leydig cell apoptosis and expression of PARP and Fas gene.Methods 20 adult healthy SD rats were randomly divided into sham-operation group and spinal cord injury (SCI) group. The animals were killed on 14 days after operation. Fluorescence activated cell sorter (FACS) was used to quantitatively assess apoptosis of Leydig cells. Western-blot was applied to detect cleaved-PARP and Fas proteins.Results On day 14 after operation, the incidence of leydig cell apoptosis in SCI group was 32.15%, significantly higher than that of sham-operation group (12.78%). Cleaved-PARP and Fas proteins were both found dramatically higher in SCI group than sham-operation group.Conclusion Spinal cord injury triggers apoptosis of Leydig cells, and PARP and Fas gene are involved in regulation of this process.
