补阳还五汤复方颗粒对缺血性脑卒中模型大鼠血浆中强啡肽和神经肽Y的影响 被引量:4

Effects of Buyang Huanwu Decoction Compound Granules on Plasma Dynorphin and Neuropeptide Y Levels in Ischemic Cerebral Stroke Rats

摘要 目的:探讨补阳还五汤复方颗粒对缺血性脑卒中模型大鼠血浆中强啡肽(dynorphin,Dyn)、神经肽Y(neuropeptide Y,NPY)的影响。方法:健康雄性SD大鼠,根据随机数字表抽取14只,作空白对照。另抽取15只,假手术处理后,作假手术组。其他大鼠称重后采用改良Pulsinelli四血管闭塞法建立临床缺血性脑卒中模型,将造模成功的大鼠纳入实验,随机分为模型组、脑心通组、补阳还五汤复方颗粒高剂量(30.4 g·kg-1)组、补阳还五汤复方颗粒低剂量(15.2 g·kg-1)组,共6个实验组。各组连续灌胃蒸馏水或相应药物7 d,用酶联免疫吸附法测定模型大鼠血浆中强啡肽,神经肽Y的含量。结果:模型组大鼠血浆中神经肽Y含量(1.31±0.05)μg·L-1与空白组(1.01±0.06)μg·L-1相比升高(P<0.01),模型组血浆中Dyn含量平均值(15.87±1.57)ng·L-1与空白组(2.31±0.95)ng·L-1相比升高(P<0.01),补阳还五汤复方颗粒组血浆中NPY含量(1.23±0.06)μg·L-1,Dyn含量(3.02±0.56)ng·L-1,与模型组相比降低,表明补阳还五汤复方颗粒能使大鼠血浆中异常升高的Dyn、NPY含量降低,趋近正常水平。结论:补阳还五汤复方颗粒对脑卒中的治疗作用可能与调节脑内神经因子水平、神经功能有关。 Objective: To explore the effects of Buyang Huanwu decoction compound granules (BYHWDCG) on the plasma dynorphin (Dyn) and neuropeptide Y (NPY) contents in ischemic cerebral stroke rats. Method: The model was reproduced by the four-vessel occlusion (4-VO) method with some modifications. The rats were treated with BYHWDCG ( 30.4, 15.2 g ·kg ^-1 ). The Dyn and NPY levels were assayed by ELISA. Result: The plasma Dyn and NPY levels in model group were increased compared with control group. However, after treated with BYHWDCG, the contents of Dyn and NPY in plasma significantly decreased. Conclusion: The BYHWDCG can protect rats with the ischemic stroke by down-regulation of neurotrophie factors.
