摘要
目的从鱿鱼皮中提取胶原蛋白并研究经化学和物理方法改性的鱿鱼皮胶原海绵的止血效果。方法从新鲜鱿鱼皮中提取得到酸溶性胶原蛋白(ASC)和酶溶性胶原蛋白(PSC),冷冻干燥制备胶原海绵,分别采用EDC交联、干热交联(DHT)、EDC/DHT结合交联这3种方法对其进行改性处理。采用兔耳创伤止血和肝脏止血试验来评价改性后胶原海绵的止血性能。结果鱿鱼皮胶原蛋白ASC和PSC为典型的I型胶原蛋白。动物止血实验结果表明ASC-E、PSC-E、ASC-E/D和PSC-E/D等4种改性鱿鱼皮胶原海绵组均能对创面起到一定止血作用,其中尤以PSC-E组的止血效果最好,优于市售明胶海绵。结论 ASC-E、PSC-E、ASC-E/D和PSC-E/D等4种改性鱿鱼皮胶原海绵组均能有效的缩短出血时间,减少出血量,达到快速止血的效果,其中PSC-E止血效果最佳。
Objective To extract collagen from squid skin and study the hemostatic effect of the collagen sponges which were modified by chemical and/or physical method.Methods Acid-soluble collagen(ASC)and pepsin-soluble collagen(PSC)from fresh squid skin were isolated and characterized.The prepared collagens were lyophilized to prepare for collagen sponges,and then crosslinked using 1-ethyl-3-(3-dimethyl aminopropyl)carbodiimide(EDC)treatment,dehydrothermal(DHT)treatment and EDC/DHT treatment,respectively.Rabbit ear wound bleeding test and liver bleeding test were measured to evaluate hemostatic properties of modified collagen sponges.Results ASC and PSC were both typical type I collagen.Results of animal hemostatic experiments showed that ASC-E,PSC-E,ASC-E/D and PSC-E/D the four groups of collagen sponges all could be a certain role in hemostasis of wound up,especially,the hemostatic effect of group PSC-E was the best and better than those of commercially available gelatin sponge.Conclusion ASC-E,PSC-E,ASC-E/D and PSC-E/D all could effectively shorten the bleeding time,decrease blood loss,increase hemostatic effect,and the hemostatic effect of PSCE was best,especially.
