两种方法测定解冻红细胞甘油残留量结果比较 被引量:2

摘要 目的通过对过碘酸钠滴定法和甘油三酯比色法测定解冻红细胞甘油残留量的结果进行比较,探讨应用甘油三酯比色法代替过碘酸钠滴定法进行解冻红细胞甘油残留量测定的可行性。方法配制不同浓度的甘油溶液,比较2种方法的精密度和准确性;测定甘油三酯比色法的线性范围、回收率、干扰试验及其他影响实验因素;用2种方法分别测定30份解冻红细胞标本的甘油残留量,统计分析二者之间的差异。结果甘油三酯比色法测定甘油含量≤2 g/L时有良好的线性关系;过碘酸钠滴定法精密度平均值为6.86,甘油三酯比色法精密度平均值为5.93;过碘酸钠滴定法准确性平均值为112.3%,甘油三酯比色法准确性平均值为97.7%;甘油三酯比色法测定甘油含量回收率平均值为99.5%;测定标本Hb浓度达到12.5 g/L时,对测定结果无干扰(t=1.646,P>0.05);标本血滤液对测定结果无显著影响(t=2.464,P>0.05);血液制剂中甘油三酯含量对测定结果无显著影响;2种方法测定30份解冻红细胞甘油残留量时二者之间差异无统计学意义(t=1.884,P>0.05)。甘油三酯比色法和过碘酸钠滴定法测定甘油残留量时二者之间具统计学意义。结论甘油三酯比色法在测定解冻红细胞甘油残留量时具有方法简单、快速、灵敏度高、稳定性好、回收率高、受干扰因素少等优点,我们认为可以代替过碘酸钠滴定法进行解冻红细胞甘油残留量的质控测定。 Objective To discuss the feasibility of replacing sodium periodate titration with triglyceride colorimetric method, by comparing the results of two methods for m.easuring glycerin residues in deglycerolized frozen-thrawed red blood ceils. Methods Glycerin solution with different concentration were prepared for comparing the degree of precision and accu- racy of two methods. The linear range of triglycerides colorimetric method, the rate of recovery, interference experiment and other factors influencing the experiment were determined. These 2 methods were used to measure glycerol residues in 30 sam- pies of thawed red blood cell, and the difference between tow methods were statistically analyzed. Results The results of tri- glyeeride colorimetric method has good linear relationship when glycerol Concentration ≤ 2 g/L. The average degree of preci- sion of sodium periodate titration and triglyceride assay was 6.86 and 5.93 respectively. The accuracy of sodium periodate ti- tration and triglyceride assay was 112.3% and 97.7% on average. The average'glycerin recovery rate measured by triglycer- ide colorimetric method was 99.5%. Hb concentration had no interference on the determination results when it reached 12.5 g/L ( t = 1. 646 ,P 〉0. 05 ). Different samples had no significant effect on results ( t = 2. 464 ,P 〉 0. 05 ), neither did blood triglyceride concentration.. The glycerin residue in 30 samples measured by 2 methods were not significantly different ( t = 1. 884,P 〉 0. 05 ). Conclusion Triglyceride colorimetric method is more simple, rapid, sensitive and stabel with high recov- ery rate, and fewer interfering factors. We conclude that we can use triglyeeride colorimetric method instead of sodium perio- date titration to measure glycerol residue in thawed erythrocytes.
