摘要
目的探讨不同浓度色素上皮衍生因子(PEDF)作用不同时间对体外培养的人肺腺癌A549细胞增殖和凋亡的影响。方法用不同浓度(0、80、160、320、640 nmol/L)的重组人PEDF蛋白对体外培养的人肺腺癌细胞株A549进行干预,在处理24、48、72 h后,分别用MTT法检测PEDF对A549细胞增殖的抑制率并比较;在作用48 h后,以流式细胞仪检测技术计算、分析A549细胞凋亡率。结果不同浓度PEDF(0、80、160、320、640 nmol/L)干预A549细胞相同作用时间(24、48、72 h)情况下,随着PEDF浓度的增加,PEDF对A549细胞的增殖抑制率递增(P<0.05);相同PEDF浓度(80、160、320、640 nmol/L)情况下,除了80、160 nmol/L PEDF作用24 h和48 h的细胞增殖抑制率间差异无统计学意义外,随着PEDF作用时间的延长,PEDF对A549细胞的增殖抑制率也基本呈递增趋势(P<0.05)。上述不同浓度的PEDF处理A549细胞48 h,细胞凋亡率随着PEDF浓度的升高呈递增趋势(P<0.05)。结论 PEDF在体外能抑制人肺腺癌细胞株A549的增殖并诱导其凋亡。PEDF抑制细胞增殖作用可能和PEDF浓度及作用时间有关,PEDF诱导细胞凋亡作用的强弱也与PEDF浓度有一定关系。
Objective To observe the effects of pigment epithelium-derived factor ( PEDF) on the proliferation and apoptosis of human lung adenocarcinoma A549 cells. Methods PEDF of various concentrations (0, 80, 160, 320 and 640 nmol/L) was used to intervene A549 cells in vitro for various time (24 h, 48 h and 72 h) . The effects of PEDF on cell proliferation was then assessed by MTT assay at the time point of 24 h, 48 h and 72 h, respectively. The rate of apoptosis induced by PEDF was determined at the time point of 48 h with flow cytometry. ResultsWith intervention by PEDF of various concentrations (0, 80, 160, 320 and 640 nmol/L), the rates of inhibition was increased by degrees with angmentation of PEDF concentration for a certain time(24,48,72 h) (P〈0.05);for a certain concentration of PEDF (80, 160, 320, or 640 nmol/L), except the rates of inhibition between 24 h and 48 h were of no statistical differences for both 80 and 160 nmol/L PEDF, the rate of inhibition was generally in-creased by degrees with augmentation of PEDF intervention time (P〈0.05). In addition, the rates of apoptosis of A549 cells induced by PEDF of various concentrations (0, 80, 160, 320 and 640 nmol/L) was increased by de-grees with augmentation of PEDF concentration ( P〈0.05 ) . Conclusion PEDF may in vitro inhibit proliferation and induce apoptosis in human lung adenocarcinoma A549 Cells. The inhibition of proliferation might be associated with PEDF concentration and intervention time, and the induction of apoptosis might also be dependent on certain concentration of PEDF.
