依布硒啉对糖尿病大鼠心脏的保护作用及其机制 被引量:2

Protective effect and mechanism of ebselen on cardiac function in diabetic rats

摘要 目的探讨抗氧化剂依布硒啉对糖尿病大鼠心脏损害的保护作用及其机制。方法采用高热量饮食联合小剂量链脲佐菌素注射的方法诱导2型糖尿病大鼠模型。造模成功后随机分为依布硒啉干预组(A组,11只)和糖尿病心肌病组(B组,10只);另取8只SD大鼠作为正常对照组(C组)。8周后检测心功能、血生化指标、心脏指数、心肌丙二醛(MDA)含量、超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH-Px)的活性。结果与C组相比,B组大鼠血糖、血脂、心脏指数升高,心功能减退,心肌MDA含量增加,SOD和GSH-PX活性降低(P<0.01);而A组各观察指标均较B组明显改善(P<0.01)。结论依布硒啉对糖尿病大鼠心脏损害具有保护作用,其机制可能与抑制氧化应激有关。 Objective To investigate the protective effect and underlying mechanism of ebselen on the cardiac damage in diabetic rats. Methods Type 2 diabetes mellitus models were induced by high-fat diet and intraperitoneal injection of streptozotocin in SD rats, which then were divided into two groups of A(treated with ebselen 50 mg/kg, 11 rats) and B(with diabetic cardiomyopathy, 10 rats). Besides, another 8 SD rats were taken as the controls (group C). After 8 weeks, the cardiac function,biochemical indicator, cardiac index, MDA content and activities of SOD and GSH-Px were detected. Results Compared with group C, the levels of blood glucose, triglyeeride, total cholesterol and MDA and cardiac index were increased(P^0. 01), while cardiac function and activities of SOD and GSH-Px were decreased in group B(P〈0. 01), which were all significantly improved in group A (P〈0. 01). Conclusion Ebselen has a protective effect on cardiac damage in diabetic rats,which may be related to the inhibition of oxidative stress.
