白细胞介素1β在顺铂诱发口腔鳞癌治疗性炎症中的作用 被引量:1

Roles of IL-1beta in therapy-induced inflammation triggered by cisplatin in oral squamous cell carcinoma

摘要 目的探讨白细胞介素1β(interleukin-1β,IL-1β)在顺铂诱发的治疗性炎症过程中的作用。方法口腔鳞状细胞癌细胞CAL27裸鼠皮下注射成瘤后随机分为两组,实验组腹腔注射顺铂进行肿瘤治疗,对照组腹腔注射生理盐水。实时定量聚合酶链式反应检测瘤体IL-1β和血管内皮生长因子(vascular endotheliel cell growth factor,VEGF)的mRNA表达,免疫组织化学检测瘤体IL-1β和VEGF的蛋白表达。结果实时定量聚合酶链式反应结果显示实验组瘤体组织IL-1β和VEGF的mRNA表达明显高于对照组;免疫组织化学检测示实验组IL-1β和VEGF蛋白表达上调,实验组与对照组IL-1β表达评分均数分别为13.10和5.84,VEGF表达评分均数分别为9.90和3.85,差异均有统计学意义(P<0.001)。结论 IL-1β参与了顺铂诱导的治疗性炎症过程,并通过上调下游效应分子VEGF的表达,造成口腔鳞癌局部炎症淤积,影响化疗效果,提示临床治疗过程中须考虑炎症因素对化疗效果的影响。 Objective This study aimed to investigate the roles of interleukin-1β (IL-113) in therapy-induced inflam- mation induced by cisplatin in xenograft model of oral squamous ceil carcinoma (OSCC). Methods Human tongue squamous cell carcinoma cell line CAL27 was inoculated into nude mice. Mice were randomly separated into 2 groups ( n = 5 ), one group were intraperitoneally injected with normal saline served as control while the other group were injected with cisplatin (2 mg/kg/4 days) for about two weeks. The expression of IL-1βand its downstream factor, vascular endo- thelial cell growth factor ( VEGF ) , in tumors were detected by real-time quantitative polymerase chain reaction ( RT- PCR) and immunehistochemistry (IHC) assay. Results Both IL-1βand VEGF expression were up-regulated in tumor tissue after treatment with cisplatin in mRNA and protein level ( P 〈 0.05 ). Conclusion IL-1β was involved in therapy- induced inflammation triggered by cisplatin. It enlarged the inflammation effect by up-regulating its downstream effector VEGF. These results suggested inflammation factors, which might alter the outcome of the chemotherapy should he aware of during clinical practice.
