尾加压素Ⅱ基因rs228651多态性与妊娠糖尿病相关性研究 被引量:1

Association between single variant rs228651 in urotensin-Ⅱand gestational diabetes mellitus

摘要 目的探讨尾加压素Ⅱ(urotensin-2,UTS-Ⅱ)基因rs228651单核苷酸多态性与妊娠糖尿病(gestational diabetes mellitus,GDM)遗传易感性的关系。方法 GDM患者120例(GDM组),正常糖耐量孕妇100例(对照组),采用限制性片段长度多态性PCR法检测2组UTS-Ⅱ基因rs228651位点基因分型,分析UTS-Ⅱ基因rs228651多态性与GDM遗传易感性的关系。结果 GDM组空腹血糖、空腹胰岛素、总胆固醇、胰岛素抵抗指数、β细胞功能指数分别为(6.05±0.51)mmol/L、14.20(13.22,15.60)mu/L,5.85(5.50,6.18)mmol/L,3.82±0.47,116.55(94.19,135.79),对照组分别为(4.40±0.40)mmol/L、10.19(9.63,10.70)mu/L,4.70(4.50,4.90)mmol/L、2.90±0.24、225.56(174.48,314.64),2组比较差异有统计学意义(P<0.05);GDM组UTS-Ⅱ基因rs228651位点GG、AA基因亚型分布频率(44.2%、15.0%)与对照组(23.0%、29.0%)比较差异有统计学意义(P<0.01);GDM组危险等位基因G分布频率(64.6%)明显高于对照组(47.0%)(P<0.01);GDM组GG、GA、AA基因亚型空腹血糖水平分别为(6.30(6.00,6.70)、5.90(5.55,6.20)、5.70(5.18,6.10)mmol/L),胰岛素抵抗指数分别为(4.04±0.35、3.72±0.47、3.43±0.10),除GG、GA亚型空腹血糖水平差异无统计学意义(P>0.05),3个亚型间空腹血糖水平和胰岛素抵抗指数比较差异均有统计学意义(P<0.05)。结论 UTS-Ⅱ基因rs228651位点(G/A)多态性与GDM易感性有关。 Objective To investigate the association between single variant rs228651 in urotensin-Ⅱ (UTS-Ⅱ ) gene and the genetic susceptibility to gestational diabetes mellitus (GDM). Methods PCR restriction fragment length polymorphism was used to detect rs228651 genotype in 120 GDM patients (GDM group) and 100 normal glucose tolerance pregnant women (control group) to analyze the relationship between the gene polymorphism and the genetic susceptibility to GDM. Results The fasting plasma glucose (FPG), fasting insulin, total cholesterol, homeostasis model assessment of insulin resistance (HOMA-IR) and HOMA-β were (6.05±0.51) mmol/L, 14.20 (13.22,15.60) mu/L, 5.85 (5.50, 6.18) mmol/L, 3.82±0.47 and 116. 55(94. 19, 135. 79) in GDM group, significantly different from those in control group ((4.40±0. 40)mmol/L, 10. 19 (9. 63, 10. 70) mu/L, 4. 70 (4. 50,4. 90) mmol/L, 2. 90±0. 24, 225. 56 (174.48, 314.64)) (P〈0.05). There were significant differences in the gene distribution frequency of GG and AA genotypes of UTS-Ⅱ rs228651 between GDM group (44.2%, 15.0%) and control group (23.0%, 29.0%) (P〈0.01). The risk allele G distribution frequency was significant higher in GDM group (64. 6%) than that in control group (47.0%) (P〈0.01). The FPG levels of GG, GA and AA genotypes were 6.30 (6.00, 6.70), 5.90 (5.55, 6.20) and 5.70 (5.18, 6.10) mmol/L and HOMA-IR were 4. 04±0. 35, 3. 72±0.47 and 3.43±0. 10 in GDM group, showing significant differences in all indexes among all genotypes (P〈0.05) except FPG level between GG and GA genotypes (P〉0.05). Conclusion Single variant rs228651 (G/A) of UTS-Ⅱ gene is associated with the genetic susceptibility to GDM.
