摘要
目的探讨糖尿病大鼠玻璃体血管内皮生长因子(vascular endothelial growth factor,VEGF)及VEGF受体2(kinase insert domain-containing receptor,KDR)水平变化与糖尿病视网膜病变的关系。方法 84只成年Wistar大鼠随机分为观察组64只和对照组20只。观察组采用腹腔注射链脲佐菌素制作糖尿病模型,对照组腹腔注射生理盐水。2组于造模当天及建模后1、2、4周测量大鼠血糖和体质量,4周后采用ELISA法检测大鼠玻璃体VEGF及KDR水平。结果观察组成功制造糖尿病大鼠模型58只,1个月后存活42只,对照组全部存活;造模成功4周时观察组大鼠体质量((297.43±33.50)g)低于对照组((374.57±43.57)g)(P<0.05);造模成功1、2、4周时观察组血糖水平分别为(27.64±5.83)、(26.36±5.12)、(25.29±5.01)mm/L,对照组分别为(5.14±1.76)、(5.36±1.73)、(5.71±1.86)mm/L,差异均有统计学意义(P<0.01);造模成功4周后观察组玻璃体VEGF及KDR水平分别为(0.288±0.026)、(2.936±0.295)μg/L,对照组分别为(0.231±0.021)、(2.394±0.227)μg/L,差异均有统计学意义(P<0.05)。结论大鼠糖尿病模型玻璃体中VEGF和KDR水平变化与糖尿病致早期视网膜病变有关。
Objective To explore the relationship of diabetic retinopathy with the levels of vitreous vascular endothelial growth factor(VEGF)and kinase insert domain-containing receptor(KDR)in diabetic rats.Methods Eighty-four adult Wistar rats were randomly divided into observation group(n=64)receiving intraperitoneal injection of streptozotocin to induce diabetes models,and control group(n=20)receiving injection of normal saline.The plasma glucose and body mass were examined in both groups on the same day,and 1,2and 4weeks after the models were established,and vitreous VEGF and KDR levels were detected by ELISA 4 weeks after the models were established.Results In observation group,58 models were successfully established and 42 rats survived one month later.Twenty rats survived in control group.The body mass was(297.43±33.50)g in observation group,significanatly lower than that in control group((374.57±43.57)g)in 4weeks after the establishment of models(P〈0.05).The plasma glucose in 1,2and 4weeks after establishment of models were(27.64±5.83),(26.36±5.12)and(25.29±5.01)mm/L in observation group,and(5.14±1.76),(5.36±1.73)and(5.71±1.86)mm/L in control group,showing significant differences between two groups(P〈0.05).The levels of VEGF and KDR were(0.288±0.026)and(2.936±0.295)μg/L in observation group,showing significant differences in comparison with control group((0.231±0.021),(2.394±0.227)μg/L)in 4weeks after the establishment of models(P〈0.05).Conclusion The changes of vitreous VEGF and KDR levels in diabetic rats are correlated with the early diabetic retinopathy.
