奶牛乳房炎性乳房链球菌GapC重组蛋白的免疫保护性研究 被引量:7

摘要 为研究奶牛乳房炎性乳房链球菌GapC蛋白的免疫保护性,本研究采用纯化后的GapC重组蛋白与免疫佐剂乳化后制备疫苗,免疫家兔后采用间接ELISA法检测抗体效价,并以小鼠为实验动物进行攻毒试验。结果显示,抗体效价高达1∶128 000,GapC蛋白免疫组对乳房链球菌的保护率较高,达70%,而全菌体免疫组的保护率仅为30%,表明GapC蛋白具有很高的免疫原性,为奶牛乳房炎的预防控制和进一步研究有效疫苗提供实验依据。 To evaluate the immune response of Streptococcus uberis GapC protein,the purified prokaryotic expressed recombinant GapC (rGapC) was emulsified with Freund's complete (for primary) and incomplete (for boost) adjuvants,respectively,which were used to stimulate the antibody production in rabbits and protection test in mice with white oil adjuvanted inactivated S.uberis as positive control.The results showed that antibody titer was up to 1:128,000 in rGapC immunized by rabbits and the rGapC also provided 70% protection rate for the vaccinated mice against the S.uberis challenge,which was higher than that of 30% protection rate for mice with the inactivated S.uberis.These data provide the evidence that the S.uberis GapC has the potential to be used as antigen for developing the subunit vaccine against S.uberis infection.
