MDC对不可分型流感嗜血杆菌P6重组蛋白免疫原性的影响

The effects of MDC as adjuvant on immunogenicity of recombinant P6 protein of nontypable Haemophilus influenzae

摘要 观察巨噬细胞源趋化因子(macrophage-derived chemokine,MDC)联合不可分型流感嗜血杆菌重组P6蛋白免疫小鼠的免疫效果,探讨MDC的佐剂效应。将50只BALB/c小鼠随机分为5组:P6重组蛋白+MDC,P6重组蛋白,MDC组,P6重组蛋白+IL-2组及PBS对照组,分别于第0 d、14 d、28 d经腹腔免疫3次,末次免疫后14 d,取小鼠血、脾标本,ELISA法检测血清中抗体IgG水平,并分析其脾淋巴细胞IL-10和IFN-γ的产生水平。CCK-8法检测各组脾淋巴细胞增殖活性。结果是小鼠血清IgG水平,P6重组蛋白联合MDC组高于PBS对照组、MDC组、P6重组蛋白+IL-2组及P6重组蛋白组(P<0.05)。P6重组蛋白联合MDC组小鼠特异性脾淋巴细胞刺激指数及其所产生的IL-10、IFN-γ水平均高于PBS对照组、MDC组及P6重组蛋白组(P<0.05)。因此MDC联合P6重组蛋白诱导小鼠的免疫应答优于P6重组蛋白单独免疫,MDC可以作为不可分型流感嗜血杆菌P6重组蛋白的佐剂。 The purpose of this study was to evaluate the adjuvant effect of macrophage-derived chemokine(MDC)on recombinant P6 protein of non-typable Haemophilus influenzae.Fifty BALB /c mice were randomly divided into five groups,they were immuned respectively by P6 recombinant protein.P6 recombinant protein plus IL-2,MDC.P6 recombinant protein+MDC and PBS.The mice of each group were immunized by intraperitoneal injection of the specific element at day 0,day 14 and day 28.Fourteen days after the last immunization,the peripheral blood and the spleen were collected to detect different immune parameters.ELISA was carried out to detect the levels of IgG,IL-10 and IFN-γ.The proliferation of spleeny lymphocytes was estimated by CCK-8 counting.In the group treated with P6 recombinant protein plus MDC,the levels of serum IgG was higher than those in the groups treated respectively with recombinant P6 plus IL-2,recombinant P6,MDC and PBS(P〈0.05),Specific lymphocyte stimulation index and levels of IL-10 and IFN-γ were also higher in the P6 recombinant protein plus MDC group than those in the other three groups(P〈0.05).Taken together,our results showed that immune responses induced by P6 recombinant protein plus MDC were superior than those induced by P6 recombinant protein alone.So,MDC may be used as an adjuvant for recombinant P6 proteins of non-typable Haemophilus influenzae.
