基质金属蛋白酶及一氧化氮合酶与脑动脉瘤形成的相关性 被引量:5

Pathophysiological roles of matrix metalloproteinases and nitric oxide synthase in cerebral aneurysm

摘要 目的 观察基质金属蛋白酶(MMP)2和9及诱导型一氧化氮合酶(iNOS)在脑动脉瘤中的表达,探讨其在脑动脉瘤病理发生中的作用.方法 取2009至2012年内蒙古自治区人民医院及北京天坛医院脑动脉瘤患者经手术切除的标本,共12例作为试验组,取10例颞叶癫痫患者经手术切除的颞叶皮层动脉标本作为对照组.应用明胶酶谱法测定标本MMP2和9的活性,化学比色法测定标本总一氧化氮合酶(TNOS)及iNOS的活性.结果 试验组MMP2和9的比活分别为(199 598±125 288)灰度面积·mg-1·L-1、(719 253±376 519)灰度面积·mg-1·L-1,均明显高于对照组(P<0.05).试验组TNOS和iNOS活力分别为(23.6±6.6)U/mg蛋白,(11.4±2.6) U/mg蛋白,TNOS试验组与对照组比较差异无统计学意义(P>0.05),而iNOS的活力与对照组比较差异有统计学意义(P<0.05),iNOS活力在TNOS活力中所占的比例两组间差异有统计学意义(P<0.05).结论 MMP2、MMP9、iNOS与脑动脉瘤的形成密切相关. Objective To explore the expressions of matrix metalloproteinases-2 (MMP-2),matrix metalloproteinases-9 (MMP-9) and inducible nitric oxide synthase (iNOS) in cerebral aneurysms,compare them with normal brain vessels tissue so as to gain a better understanding of the pathogenesis of cerebral aneurysms.Methods Twelve samples of cerebral aneurysms were obtained during operations and 10 cortical arteries as controls during surgery for temporal lobe epilepsy from 2009 to 2012 at Inner Mongolia People's Hospital and Beijing Tiantan Hospital.The activities of MMP-2,MMP-9 and iNOS in specimens were detected with spectrophotometry and substrate gel zymography.Results The MMP-2 and MMP-9 levels in cerebral aneurysm group were (199 598 ± 125 288) gray scale area · mg-1 · L-1 and (719 253 ± 376 519) gray scale area · mg-1 · L-1.Both in cerebral aneurysm group were significantly higher than that in control group(P 〈0.05).The TNOS and iNOS levels in cerebral aneurysm group were(23.6 ± 6.6) and (11.4 ± 2.6) U/mgprot.The difference of TNOS level was not significant between aneurysm and control groups (P 〉0.05) while the levels of iNOS and iNOS/TNOS in cerebral aneurysm group were significantly higher than that in control group (P 〈 0.05).Conclusion MMP-2,MMP-9 and iNOS are closely correlated with cerebral aneurysm.
