摘要
目的:通过甲基化抑制剂5-氮杂-2'-脱氧胞苷(5-Aza-CdR)作用于乳腺癌细胞株MCF-7,探索5-Aza-CdR逆转乳腺癌抑癌基因RASSF1A甲基化作用的机理。方法:体外培养乳腺癌细胞株MCF-7细胞,使用不同浓度5-氮杂-2'-脱氧胞苷处理MCF-7细胞,对照组不加药。MSP及Western blot分别检测经5-Aza-CdR处理前、后RASSF1A DNA、蛋白的表达情况。结果:经去甲基化药5-Aza-CdR作用后,实验组恢复了RASSF1A的表达,明显高于对照组(P<0.05)。结论:去甲基化药5-Aza-CdR可以逆转乳腺癌细胞RASSF1A的甲基化作用,恢复了RASSF1A蛋白的表达,为临床治疗乳腺癌提供了理论基础。
Objective:In order to explore RASSF1A effectiveness in breast cancer,this experiment detects the ex-pression of RASSF1A DNA and protein before and after methyltransferases inhibitor(5-Aza-CdR)used in breast cancer cell strain MCF-7. Methods:Breast cancer cell lines MCF-7 were treated with 5 and 10μmol/L 5-Aza-CdR,respectively. The expression of tumor suppressor gene RASSF1 DNA,protein was detected by methylation speci-ficity PCR and Western blot. Results:RASSF1A DNA and RASSF1A protein were expressed after 5-Aza-CdR used in breast cancer cells,but they were not expressed before 5 -Aza -CdR used. Conclusion:RASSF1A methylated completely in breast cancer cell line MCF-7,5-Aza-CdR can reverse methylation,make RASSF1A gene express again. The experiment provides basis of 5-Aza-CdRˊs application in clinically treating breast cancer.
